ObjectiveTo investigate the levels of angiopoietin-2 (Ang-2) in the vitreous fluids of patients with proliferative diabetic retinopathy (PDR) and to ascertain their involvement, if any, in angiogenesis of PDR. To observe the ransform of morph and expression of Ang-2 in human umbilical vein endothelial cells(HUVECs), which was culturaled in different condition. To approach the contribution of Ang-2 in PDR.Materials and Methods1. Twenty-six eyes of 25 patients with PDR and 5 eyes of 5 persons without eye diseases (control group) vitreous fluid samples were obtained at vitrectomy, and the levels of Ang-2 were measured by enzyme-linked immunosorbent assay. Kruskal-Wallis test and non-parametric (Spearman) correlation tests were used to analyse the data.2. To prepare cultural HUVECs slides, HUVECs slides were divided into three groups at random: A group was blank group, B group was test control group(given vitreous fluids of health adult), C group was test group (given vitreous fluids of PDR patient). After raises for 24 hours, all the slides were fixed and done Immunocytochemical stain of Ang-2.Results1. Ang-2 concentrations in the vitreous fluids of patients with PDRV(540.5470pg/ml) and PDRⅥ(472.2060 pg/ml) were obviously higher than that in health adult(7.9170 pg/ml)(P<0.01). 2. Ang-2 concentrations with the patient history of the vision aggravation were negative correlation(r=—0.802, P<0.01).3. Ang-2 concentrations with the patient history of the vision descent were non-correlation(P>0.05).4. Ang-2 concentrations with the patient history of diabetes mellitus were noncorrelation(P>0.05).5. The HUVECs treated with vitreous fluids of PDR patients showed crossing-shaped or anomal-distribution, anomal-tubiform structure can be seen, the cell is fusiform shape, and Ang-2 immunocytochemical staining showed strong positive expression. While treated with nomal vitreous fluids, the HUVECs distributed equally, more regular tubiform structure can be seen, and the Ang-2 showed weak positive expression.ConclusionAng-2 concentrations in the vitreous fluids of patients PDR were obviously higher than that in health adult. These data suggest an association of Ang-2 with angiogenic activity in PDR, Ang-2 may be one of the initiation factors of retina neovascularization.
