Objetives: To investigate the expression of IGF-1 and its relation to the angiogenesis,activity of cellproliferation in human meningiomas.To explore the role of IGF-1 in the occurrence and development of meningiomas.To provide the theoretical and experimental foundation further for clinical therapy of human meningiomas.Methods: 42 samples of meningiomas and 7 samples of normal dura were collected in the operations,Which fixted by 10% neutral formaldehyde and then desiccation,dipped into wax and embeded routinely.All sections of sampals were stained with HE.And then four groups were divided into,respectively are normal dura group,WHO I grade,II grade,III grade meningioma groups. Immunohistochemistry stains(S-P) were performed to detect the expression intensity of IGF-1 in 42 samples of meningiomas and 7 samples of normal dura.The calculation method of expression intensity is that each sample were selected 10 high-power fields(×400) and calculated the positive cell of 1000 tumor cells.The data expressed with percentage. Immunohistochemistry stains(S-P) were performed to detect the expression of FVIII- RAg(polyclonal antibody) and display the tumor micro- vessel.Tumour angiogenesis was assa- yed by the value of MVD. First of all,observe the tissue section overall and choose the highest regions of microvessel density at low-power fields(×100),and then observe at high-power fields(×400),an endothelial cell or a group of endothelial cells was stained into deep yellow as a microvessel.Record the num- ble of microvessel at 5 high-power fields(×400),the average of the numbles as the MVD value of each sample. Immunohistoch- emistry stains(S-P) were performed to detect the expression in- tensity of PCNA in the selected samples of meningiomas, act- ivity of cellproliferation in meningiomas expressed with PCNA labelling index(PCNA-LI). The calculation method of its expression intensity is that each sample were selected 10 unduplicated high-power fields(×400) and calculated the positive cell of 100 tumor cells in each high-power fields(×400)(vascular endothelial cell, lymphocyte and other cells excluded),calculated the ratio of positive cell of the tumor cells as the PCNA-LI,the data expressed with percentage. Applying the SPSS 11.5 software for the statistical analysis of the data of the experiment outcome,the date which calculated were expressed with mean±standard deviation( x±s).P value less than 0.05 was considered as statistically significant.Results:1.Result of the pathological grade of the meningiomas:All samples of the meningiomas stained with the way of HE and then confirmation the pathological diagnosis according to the newest WHO(2000) CNS tumor classified standard.Among of it,26 examples are WHO I,6 examples are WHO II,10 examples are WHO III.2.Result of immunohistochemistry stains(S-P) of IGF-1: The high levels of IGF-1 expression were detected in 31 of 42 in the the meningiomas,the positive rate was 73.8%.The positive cells of IGF-1 displayed that yellow and deep yellow were stained in cytoplasm of the tumor cells, distributed like particles and cluster.No stained in negative contrast and negative staining in normal dura.In WHO I grade,15 of 26 were detected, the positive rate of IGF-1 expression was 57.6%.The IGF-1 positive cells were detected in all WHO II and WHO III grade tissue section of the meningiomas.Intensity of IGF-1 expression in each group were not the same,respectively 0%,(15.7±12.8)%,(47.2±11.7)%和(62.9±12.9)%.Intensity of IGF-1 expression increased accompany with the tumor grade raised.WHO III grade meningioma group compared with other groups,WHO II grade group compared with WHO I and Normal Dura groups,WHO I grade group compared with Normal Dura group,and the difference have statistics significance(q test,P <0.05).3.Result of immunohistochemistry stains(S-P) of FVIII- RAg:The positive cells which the microvessel endothelial cells of FVIII-RAg expression were stained into deep yell- ow,microvessels density were not equal distribution at the different spot but the same tumor,microvessels were relative intensive in some areas.FVIII-Ag expression in all samples,the value of MVD respectively 5.3±0.9,16.2±6.6,21.2±6.4和27.6±6.3 in group of Normal Dura,WHO I,WHO II and WHO III.The value of MVD increased according to the tumor grade raised.WHO III compared with other groups,WHO II and WHO I group compared with Normal Dura group,the difference have statistics significance(q test,P <0.05).But WHO I compared with WHO II, the difference have not statistics significance(q test,P>0.05).4.Result of immunohistochemistry stains(S-P) of PCNA: The positive cells which the tumor cells of PCNA expression were stained into deep yellow in nucleus,staining intensity existed obviously difference at the same section .Take on heterogenous distribution. Negative staining in normal dura. WHO I grade,22 of 26 were detected, the positive rate of PCNA expression was 84.62%.The PCNA positive cells were detected in all WHO II and WHO III grade tissue section of the meningiomas.PCNA-LI respectively 0%,(9.6±5.4)%,(30.6±5.6)%和(48.7±15.1)% in group of Normal Dura,WHO I,WHO II and WHO III. PCNA-LI increased according to the tumor grade raised.Four groups compared with each other,the difference all have statistics significance(q test,P <0.05).5.Result of correlation analysis displayed: IGF-1,MVD and PCNA are positive correlation each other.Conclusion:1. The high levels of IGF-1 expression existed in the meningiomas.IGF-1 facilitated the occurrence and development of meningiomas by the way of autocrine or para- crine loop,provided a new target for gene therapy meningioma.2.Intensity of IGF-1 expression relate to the angiogenesis of meningioma,have the stimulation of meningioma angiogene- sis,provide the theoretical and experimental foundation further for clinical anti-angiogenesis therapy of meningiomas.3. Intensity of IGF-1 expression relate to the activity of cellproliferation of meningioma,it can be used the reference marker to evaluate pathological grade.
