Cisplatin is a antineoplastic agent with remarkable curative effect against cancer, but there are many side effects. Cisplatin is a kind of frequently-used clinic chemotherapeutics in our country.Cisplatin can cause many side effects, nephrotoxicity is the most serious one, which has influenced curative effect and clinic use of cisplatin seriously.Mechanism of cisplatin-induced nephrotoxicity maybe relate to oxygen free radical which produces by cisplatin in organism. At present, the methods of reducing cisplatin-induced nephrotoxicity are composing and selecting some compositions include platinum or other platinum metals, treating a case of cancer with cisplatin and other drugs at the same time. N-MG, amifostine, ginsenoside, ganoderma lucidum polysaccharide are reported. There are many biological activities of ganoderma lucidum polysaccharide, such as anticancer action, immunoregulation, radioresistance, anti-oxidation, etc.Because of so many biological activities of ganoderma lucidum polysaccharide, this experiment use ganoderma lucidum polysaccharide that distilled by our lab and biochemistry and molecular biology lab of our college, to study prevention function of ganoderma lucidum polysaccharide on cisplatin-induced nephrotoxicity. The experiment includes making tissue slices, mensurating GSH-Px, MDA, SOD.Results:1. On the HE slices of control group, celluar welling and karyometry didn't appeare. On the HE slices of GLP group, celluar welling and karyometry are rare. On the CDDP group's HE slices, celluar welling, karyometry , cells aggregation to edge hyperchromatic cells were observed usually, renal tubules vacuolization are rare. On the CDDP+GLP group's slices, celluar welling and karyometry are rare, renal tubules vacuolization didn't appeare.2. GSH-PX was measured, compared to the control group, the GSH-PX enzyme activity of CDDP group, GLP group and CDDP+GLP group reduced obviously. The numerical value of GLP group's GSH-PX is bigger than CDDP group. CDDP+GLP group's GSH-PX is bigger than CDDP group a little.3. The content of MDA was measured, compared to the control group, the content of MDA increased obviously, GLP group's content of MDA increased obviously too. GLP group's content of MDA is much higher than CDDP+GLP group's. CDDP+GLP group's content of MDA is much lower than CDDP group's.4. SOD was measured, compared to the control group, CDDP group's SOD reduced, GLP group's SOD increased obviously. GLP group's SOD is much higher than CDDP group's. CDDP+GLP group's SOD is much higher than CDDP group's.Concludes:The ganoderma lucidum polysaccharide can increases SOD activity, reduces the content of MDA, elevate GSH-Px enzyme activity a little. In HE slices, celluar welling and karyometry reduced renal tubules vacuolization remitted. We can say this kind of ganoderma lucidum polysaccharide can prevent cisplatin-induced nephrotoxicity, maybe relates to that ganoderma lucidum polysaccharide can eliminate OFR and lessen lipid peroxidation.
