Expression Of Survivin, COX-2 And Bcl-2 In Non-Hodgkin's Lymphoma And Their Relationship

Objective: In this study we detected the expression of COX-2,Survivin and Bcl-2 in non-Hodgin's lymphoma (NHL) and studied their relationship.Materials and Methods: Forty-four cases of NHL and twenty cases of reactive lymphoid hyperplasia(RH) tissues taken for routine diagnosis were collected from our hospital.These cases were diagnosed between January 2005 and May 2007.Immunohistochemistry MaxVisionTM staining was adopted to detect the expression rates of COX-2,Survivin and Bcl-2 protein in the paraffin wax samples of NHL and RH.We did the statistics analysis with x2 test,Fisher exact probabilities and rank corrlation. P<0.05 has statistic significance.Results:1.The positive expression rate of COX-2,Survivin,Bcl-2 in RH was 40.00%,40.00%,20.00% respectively,in NHL was 68.18%,70.45%,63.64%. There was significant difference in the expression rates of them between NHL and RH.2.COX-2 protein and Survivin protein were positively coexpressed in 24 cases NHL,nagetively coexpressed in 7 cases NHL. There was positive correlation between the expression of COX-2 and Survivin protein in NHL (p<0.05,r=0.306); COX-2 protein and Bcl-2 protein were positively coexpressed in 18 cases NHL,nagetively coexpressed in 4 cases NHL.There was no correlation between the expression of COX-2 and Bcl-2 protein in NHL(p>0.05,r=-0.111);Survivin protein and Bcl-2 protein were positively coexpressed in 23 cases NHL,nagetively coexpressed in 8 cases NHL .There was positive correlation between the expression of Survivin and Bcl-2 protein in NHL(p<0.05,r=0.306).3.There was no correlation between the expression of COX-2,Survivin and Bcl-2 protein with age,sex,LDH,extra-nodal extension,IPI score and B symptoms,between the expression of Bcl-2 and Survivin protein with Ann Arbor stage,and between the expression of COX-2 protein with clinic group,but there were significant differences in the expression of COX-2 protein between stageâ… +â…¡and stageâ…¢+â…£,and in the expression of Survivin and Bcl-2 protein between indolent lymphoma groups and aggressive lymphoma groups.4.The positive expression rate of Bcl-2 in T-NHL was 33.33% ,in B-NHL was 75.00%. There was a significant difference between them(P<0.05).Conclution:1.There were significant differences in the expression of COX-2,Survivin and Bcl-2 protein between NHL and RH,so to detect the expression of them has clinical value to discriminate the two diseases.2. There were significant differences in the expression of Survivin and Bcl-2 protein between indolent lymphoma groups and aggressive lymphoma groups,so to detect the expression of them has clinical value to estimate the malignant degress of lymphoma.3.There was a significant difference between the positive expression of COX-2 and clinic stage. Stageâ…¢+â…£higer than stageâ… +â…¡,which demonstrates that there was a correlation between COX-2 protein expression and stages of NHL. To detect the expression of it could predict the prognosis for the patients with NHL and guide clinical therapy.4.There was a positive correlation between the expression of COX-2 and Survivin protein,between the expression of Survivin and Bcl-2 protein,which indicates that they may play a synergistic role in the occurrence and development of NHL.5.There was a significant difference between the expression of Bcl-2 in T-NHL and B-NHL.
