Objective: To study the expression of IL-6, IL-18 and coagulation factorâ…§-related antigen (Fâ…§RAg) of the renal tissue and the effect of anisodamine in overtraining-induced acute kidney injury (OTIAKI) rats, explore the mechanism of OTIAKI , and provide new theory method for clinical treatment.Methods: 1.Group Thirty six male Wistar rats were randomly divided into 6 group: sedentary control group (CN), immediately after exhaustive swimming group (ESI), 6 hours after exhaustive swimming group (ES 6h), 24 hours after exhaustive swimming group (ES 24h), 6 hours after exhaustive swimming and received a single intra-abdomen injection of anisodamine at a dose of 10mg/kg at 20 minutes before swimming group (AD 6h), 24 hours after exhaustive swimming and received a single intra-abdomen injection of anisodamine at a dose of 10mg/kg at 20 minutes before swimming group (AD 24h) 2.Developing animal model Rats of each group were given adapt swimming 30 minutes at the first day, on the second day, they were put into the water till exhausted. 3. Collecting samples. Rats of each group were anaesthetized by inhaling ether. The left kidney of rats were removed and saved in formaldehyde solution in order to observe histological and immunohistochemical staining changes. The right kidney were removed and saved in liquid nitrogen for detecting IL-6, IL-18 by ELISA method. 4. The expression of IL-6, IL-18 were detected by ELISA method, the Fâ…§RAG by immunohistochemical staining. The results of immunohistochemical staining were analyzed with the pathological image analysis system. 5. All data were finished by SPSS 12.0, and were deemed having statistical significance when (P<0.05).Result: 1.There was only slight expression of IL-6 in kidney of CN group rats, while was increased in ESI group (P<0.05). Compared with group CN, the expression of IL-6 in kidney had no significant difference in ES 6h, ES 24h, AD 6h, AD 24h group (P>0.05). 2. There was only slight expression of IL-18 in kidney of CN group rats, the expression increased in ESI group (P<0.01), and reached a higher level in ES 6h group(P<0.05), then decreased in ES 24h group, but also higher compared with CN group(P<0.05). In AD 6h group and AD 24h group, the expression of IL-18 in kidney of rats decreased. 3. There was a moderate expression of Fâ…§RAG in kidney of rats in CN group, the expression increased in ES 6h group(P<0.05), reach the a higher level in ES 24h group. Compared with ES group, the expression of Fâ…§RAG in kidney of rats increased in AD 6h group and AD 24h group.Conclusions: 1. The expression of IL-6, IL-18 of the renal tissue was increased in ESI group, the expression of IL-18 was higher compared with ESI group and the expression of IL-6 was decreased to CN group level in ES 6h group, and the expression of IL-6 and IL-18 decreased in ES 24h group. These results suggested that inflammatory response play a key role early in OTIAKI. 2. The expression of Fâ…§RAG increased in ES6h group, it reached a higher level in ES24h group. These results showed that the renal blood flow could be increased by the hyperplasma of capillary vessel after exhaustive swimming, which could be the important mechanism of renal tissue cell apoptosis rather than necrosis. 3. Anisodamine can decrease the degree of inflammatory response in renal, increase density of capillary vessel, has protective effect on OTIAKI.
