Objective: To investigate the effects of Yiqi Huoxue Huayu (YHH) method on the expression of c-myc and N-ras mRNA in precancerous lesions of liver cancer, and reveal the mechanism of YHH method in the treatment of hepatic precancerous lesion on the level of molecular biology.Methods: One hundred and sixty Wistar rats were randomly divided into normal control group, model group,high-dose group of YHH decoction (YHH I group) and low-dose group of YHH decoction (YHHâ…¡group), 40 of one group. During the experiment, YHH decoction was administered to interfere the hepatocarcinogenesis induced with DEN in rats. At 4wk,8wk,12wk and 16wk,the expression of c-myc and N-ras mRNA of hepatocyte were detected by applying RT-PCR technology and the pathological changes in liver tissue was observed by light microscope.Results: In the different stages of the experiment,compared with that in the model group,pathological changes in liver tissue of YHHâ… group and YHHâ…¡group have obviously improved;The expression of c-myc and N-ras mRNA of hepatocyte in the model group was much higher than that in the normal control group(P<0.05). At 8wk,12wk and 16wk,the expression of c-myc and N-ras mRNA of YHHâ… group and YHHâ…¡group was significantly different from that in the model group(P<0.05).Compared with that in the YHHâ…¡group,the expression of c-myc and N-ras mRNA of YHHâ… group had distinctly statistical difference (P<0.05).Conclusion: YHH method can effectively inhibite the precancerous lesions of liver cancer in rats,the mechanism of treatment may be relative to regulate expression of c-myc and N-ras.
