| Objective:To study the effect of Yiqi Yangyin Huoxueprinciple on the TGF-β/Smad signal transduction pathway in ratswith precancerous lesions of gastric carcinoma.Method:75 Wistar rats were randomly divided into modelgroup,normal control group,low dose group,high dose groupand Weimeisu group of 15.The rat model of precancerous lesionsof gastric carcinoma(PLGC) was established by four pathogenicfactors in which MNNG was mainly used.Weimeisu was given1.0g/kgper day inweimeisu group;YiqiYangyinHuoxueRecipeof 1.0g/ml,2.0g/ml drug concentration were given 10ml/kg perday in low dose group and high dose group.After 16 weeks,toobserve the pathological changes in rat gastric mucosa as well asTGF-β1,Sma4 mRNA expression and ERK1 levels.Result:(1) The expression of TGF-β1 in model group wassignificantly increased compared with normal control group.Theexpression of TGF-β1 in low dose group and high dose group wasreduced compared with the model group,and the differencesbetween groups was statistically significant(X~2 = 59.671;P＜0.05);(2) The expression of Smad4 in model group wassignificantly reduced compared with normal control group.Theexpression of Smad4 in low dose group and high dose group wasincreased compared with the model group,and the differencesbetween groups was statistically significant(F=433.572;P＜0.05);(3) The level of ERK1 in model group was significantlyincreased compared with normal control group.The level ofERK1 in low dose group and high dose group was reducedcompared with the model group,and the differences betweengroups was statistically significant(F=5.441;P＜0.05);(4) There is significantly statistical relationship between each other ofTGF-β1,Smad4 and ERK1(P＜0.05).Conclusion:Yiqi Yangyin Huoxue principle can improve thepathological conditions of gastric mucosa in rat with PLGC。Andit can also prevent the progress of PLGC by reducing TGF-β1mRNA expression and ERK1 levels,increasing the expression ofSmad4 mRNA.
