ObjectiveSmoking and passive smoking impact on testicular function, semen quality and reproductive endocrine function on male reproductive system toxicity,long-term low-intensity endurance exercise affect the body but have a good role in many aspects.The experiment observe nine weeks low-intensity endurance training impact testicular tissue structure,sperm motility,serum testosterone concentration in rats with passive smoking,determine the antioxidant enzyme,NOS,LDH,SDH,ACP and AKP activity changes on testicular tissue to explore the low-intensity endurance exercise effect on the damage of second-hand smoke to the reproductive system and the repair mechanisms for the application to reduce smoking damage and provide experimental evidence for passive smoking.Research MethodsHealthy adult male SD rats with 40(purchased in Hunan Agricultural University Animal Center),were randomly divided into 4 groups according to body weight:control group(C group), smoking group(S group),exercise group(E group) and the sports and smoking group(ES group),10 rats in each group.C group was not training,not to passive smoking;E and ES groups trained with low-intensity endurance train for nine weeks,six days a week,the intensity is 18m/min,rats exercise for 10 mins on the first week,exercise 60 mins on the sixth week and the last three weeks;S and ES groups passive smoked for 9 weeks,6 days a week,20 cigarettes one day.Weighing on the experimental rats, weigh the testis and epididymis weight after sacrificed, calculated organ coefficient;check routine blood tests;testis fixed,paraffin-embedded sections after dehydration staining,observate histopathological changes on light microscopy;microscope more than 200 sperm,recommend sperm motility according to WHO standards;observe the testosterone value of serum and testicular tissues with chemical immunoassay; use colorimetric determination observe serum and testicular tissue malondialdehyde,nitric oxide formation and activity of superoxide dismutase,glutathione peroxidase,nitric oxide synthase,lactate dehydrogenase,succinate dehydrogenase,acid phosphatase and alkaline phosphatase activity.Use analysis of variance for the measured values of the statistical,indicate statistical analysis,obtaine experimental results.Findings1)Compared with the C group,S group,E group and the ES group rats body weight and testis and epididymis organ coefficient have non-significant difference(P>0.05);ES group sperm activity significantly decreased(P<0.05),S group sperm activity was significantly lower(P<0.01);serum and organizational testosterone values of E group significantly increased(P<0.05),S group,ES group were no significant change (P>0.05);testis structure of E group was normal,ES rats testis all levels spermatogenic cells and sperm cells reduction,S group all levels spermatogenic cells,sperm cells and sperm decreased significantly.2)Compared with the C group,serum MDA significantly decreased,GSH-Px activity significantly increased(P<0.05) of E group,serum SOD,serum and tissues GSH-Px activity of S group was significantly lower(P<0.01),serum and testicular tissue MDA was significantly higher(P<0.01);testicular tissue MDA content,SOD activity of ES group was significantly higher (P<0.05),serum MDA content,SOD,GSH-Px activity was no significant change(P>0.05).3) Compared with the C group,NOS activity in serum of S group was significantly higher(P<0.01),ES group was significantly higher(P<0.05);serum NO generation of S group,E group was significantly higher(P<0.05),ES group was no significant difference(P>0.05);NOS activity in testicular tissue,NO generation of S group was significantly higher(P<0.05),E group,ES group was no significant difference(P>0.05).4) Compared with the C group,the serum LDH activity of S group was significantly lower(P<0.05),E group,ES group was no significant difference(P>0.05);testis LDH activity of S group was significantly lower(P<0.05),there were not significant differences in E group,ES group(P>0.05).Serum SDH activity of S group,E group and the ES group was no significant change(P>0.05);testis SDH activity of S group was significantly lower(P<0.05),E group,ES group was no significant difference between groups(P>0.05).Serum and testicular tissue ACP,AKP activity in E group,S group and the ES group was no significant difference(P>0.05).Conclusion1) Passive smoking damage to the testis organizational structure,decreased sperm motility;nine weeks of low-intensity endurance exercise increase serum and testis testosterone value;low-intensity endurance exercise reduce the damage of passive smoking on the testicular structure and sperm.2) Passive smoking reduced serum SOD,GSH-Px activity, increased serum and testicular tissue MDA content; low-intensity endurahce exercise can increase serum and testicular tissue GSH-Px activity and reduced MDA generation; low-intensity endurance exercise reduce serum MDA content because of passive smoking,reduce passive smoking oxidative damage in rat testicular tissues.3) Passive smoking made serum testicular tissue NOS activity, NO generation increase;low-intensity endurance exercise effect was not obvious;interaction of low-intensity endurance exercise and smoking increased serum stability of testicular tissue NOS activity,slow NO generation increased,reduce the excessive NO generated damage.4) Passive smoking made testicular tissues serum LDH,SDH activity decrease;low-intensity endurance exercise made serum and testicular tissue LDH activity,serum SDH,ACP and AKP activity increased;low-intensity endurance exercise made passive smoking serum and testicular tissue SDH,LDH,ACP,AKP activity stabilized at a certain level,reduce tobacco smoke damage on supporting cells and testicular tissue stromal cells and its energy metabolism.
