Objective:We duplicated the hyperlipemia Mice model with high-fat forage, we interfered the Mice with Acupuncture of jianpi tongfu and xiezhuo. And we studied the action mechanism of this formula in regulating lipid metabolism through observing its effect on TC,TG,HDL-C,LDL-C; We also studied its action mechanism in resisting lipid peroxidation injury through observing its effect on MDA,SOD; We studied the action mechanism of this formula in vasomotion state and blood flow through observing its effect on ET, NO.Methods:Part I Effect of Acupuncture of jianpi tongfu and xiezhuo on lipid metabolism of hyperlipemia mice60 healthy male KM mice were divided into 4 groups at random after fed adaptive one week,namely, (1) Normal control group20 (normal group); (2) Model control group20 (model group);(3)Acupuncture group10;(4)Simvastatin control group 10 (simvastatin group). Except for normal group was fed with ordinary forage, the rest of groups were all fed with high-fat forage.The 10 mice of normal and model group were decapitated and their blood were drawn out at the first 15 days to measure the TC and TG in order to Look at the success of model.At the same time,the Acupuncture group and simvastatin group begin the treatment,after fixed the mice of Acupuncture group in test-bed,we acupuncture the points of fenglong,sanyinjiao and quchi and added Korean Acupuncture Instrument on fenglong and quchi,Waveform: Density wave, Frequency2Hz,Strength conditioning as Limbs trembled slightly and can be tolerated for the degree,noce every day,10 minutes noce.the treatment lasted for 10 days. Mice in simvastatin group were given stomach-perfusion once a day and every time the drug volume was calculated by 0.2 ml/10g. Stomach-perfusion lasted for 10 days. Normal,model and Acupuncture group were given with same volume distilled water.The rest mice of other gourp were fixed in test-bed same as the Acupuncture group. Then 12 hours after fasting and not prohibiting water of the last time, the mice were decapitated and their blood were drawn out, the blood sample was made centrifugation with low temperature and the blood serum was isolated. It was sealed up and keep temperature of under -20℃to examine them later. Finally the contents of TC,TG,HDL-C,LDL-C were measured with biochemistry method.Part II Effect of Acupuncture of jianpi tongfu and xiezhuo on the content of MDA and activity of SOD in serum of hyperlipemia mice40 healthy male KM mice were divided into 4 groups at random after fed adaptive one week,namely, (1)Normal group, (2) Model group,(3) Acupuncture group,(4) Simvastatin group, Every group 10.The duplication of the hyperlipemia animal model, acupuncture and drug-given were exactly same as the part one. After 10 days continuous acupuncture and stomach-perfusion,then 12 hours after fasting and not prohibiting water of the last time, the mice were decapitated and their blood were drawn out, the blood sample was made centrifugation with low temperature and the blood serum was isolated. It was sealed up and keep temperature of under -20℃to examine them later. The content of MDA was examined by the method of TBA colorimeter analysis and activity of SOD was examined by the method of xanthine oxidase.Part III Effect of Acupuncture of jianpi tongfu and xiezhuo on the contents of ET and NO in serum of hyperlipemia miceThe group distribution was exactly same as the part one, and the duplication of the hyperlipemia animal model, acupuncture and drug-given were exactly same as the part one. It was sealed up and keep temperature of under -20℃to examine them later.The content of ET was examined by the method of Radioimmunoassay and The content of NO was examined by the method of Nitrate reductase.Results:Part I Effect of Acupuncture of jianpi tongfu and xiezhuo on lipid metabolism of in serum hyperlipemia mice1 The contents Comparison of TG,TC,HDL-C,LDL-C in serum of normal and Model groupThe contents of TC,TG,LDL-C in serum of model group (5.72±0.85,1.31±0.32,2.68±0.55) mmol/L were all higher than normal group (3.08±0.74, 0.70±0.21, 1.83±0.36) mmol/L (P<0.01),The content of HDL-C in serum of model group (0.60±0.26) mmol/L was lower than normal group (0.94±0.30) mmol/L (P<0.05), Show that the model successfully.2 Effect of Acupuncture of jianpi tongfu and xiezhuo on the contents of TC, TG, HDL-C and LDL-C in serum of hyperlipemia miceThe contents of TC, TG, LDL-C in serum of model group (5.83±1.07,1.25±0.35,2.59±0.59) mmol/L were all higher than normal group (3.26±0.71,0.74±0.18,1.83±0.42) mmol/L (P<0.01),while HDL-C of model group (0.61±0.27)mmol/L was lower than normal group(0.95±0.34)mmol/L(P < 0.01); the contents of TC,TG,LDL-C in serum of Acupuncture and simvastatin group were all lower than the model group, while HDL-C was higher than the model group (P<0.05 or P<0.01); the contents of TC,LDL-C in serum of Acupuncture group(3.33±0.77,1.55±0.27)mmol/L were all lower significantly than simvastatin group(4.36±0.82,2.08±0.28)mmol/L(P<0.05 or P<0.01). And there was no significant difference in the contents of TG, HDL-C between Acupuncture group (0.81±0.10,0.92±0.19)mmol/L and simvastatin group (0.78±0.21,0.93±0.24)mmol/L (P>0.05).Part II Effect of Acupuncture of jianpi tongfu and xiezhuo on the content of MDA and activity of SOD in serum of hyperlipemia mice1 Effect of Acupuncture of jianpi tongfu and xiezhuo on the content of MDA in serum of hyperlipemia miceThe content of MDA in serum of model group(29.94±2.79)nmol/L was higher significantly than normal group(23.33±3.44)nmol/L (P<0.01), the contents of MDA in serum of Acupuncture and simvastatin group were all lower than the model group(P < 0.01), And there was no significant difference in the content of MDA between Acupuncture group(21.58±3.54)nmol/L and simvastatin group (23.57±2.61)nmol/L (P>0.05).2 Effect of Acupuncture of jianpi tongfu and xiezhuo on the activity of SOD in serum of hyperlipemia miceThe content of SOD in serum of model group(222.66±25.48)u/ml was lower significantly than normal group(266.00±15.05)u/ml (P<0.01), the contents of SOD in serum of Acupuncture and simvastatin group were all higher than the model group(P<0.01 or P<0.05),and the content of SOD in serum of Acupuncture group(264.57±14.46)u/ml was higher than the simvastatin group(244.12±26.75)u/ml(P<0.05). Part III Effect of Acupuncture of jianpi tongfu and xiezhuo on the contents of ET and NO in serum of hyperlipemia mice1 Effect of Acupuncture of jianpi tongfu and xiezhuo on the content of ET in serum of hyperlipemia miceThe content of ET in serum of model group(67.07±11.98)Pg/ml was higher significantly than normal group(45.50±8.79)Pg/ml (P<0.01), the contents of ET in serum of Acupuncture and simvastatin group were all lower than the model group(P<0.01 or P<0.05), and the content of ET in serum of Acupuncture group(44.51±5.23)Pg/ml was lower than the simvastatin group(56.21±12.18)Pg/ml (P<0.05).2 Effect of Acupuncture of jianpi tongfu and xiezhuo on the contents of NO in serum of hyperlipemia miceThe content of NO in serum of model group(63.83±9.19)μmol/L was lower significantly than normal group(87.26±6.10)μmol/L(P<0.01), the contents of NO in serum of Acupuncture and simvastatin group were all higher than the model group(P<0.01),and the content of NO in serum of Acupuncture group(85.02±10.06)μmol/L was higher than the simvastatin group(75.80±9.11)μmol/L (P<0.05).Conclusion:1 Acupuncture of jianpi tongfu and xiezhuo has decreased the contents of TC,TG,LDL-C and increased the content of HDL-C in serum. The result shows that it has marked function of regulating lipid metabolism.2 It obviously increased the activity of SOD, and decreased the content of MDA in serum. The result shows that it has marked function of inhibiting lipid peroxidation reaction and raising anti-oxidative ability. It could reduce oxidative damage of hyperlipemia through amending free radical metabolism.3 It obviously decreased the content of ET and increases the content of NO in serum. The results shows that it has marked function of regulating the normal contraction and dilation state of vascular smooth muscle, which in order to remove the platelet coagulation with thrombosis. It could prevent the blood vessel endothelium damage of hyperlipemia and protect blood vessel endothelium function.4 The acupuncture's action of decreasing TC,LDL-C,ET and increasing SOD,NO is better than Simvastatin obviously, at the same time to improve blood lipids, but also has significantly enhanced the antioxidant capacity and protection of vascular endothelial function.
