Objective:Whether the addition of Fructose-1,6-diphosphate(FDP)and Salvia Miltiorrhiza extract(SM)to St.Thomas solution could improve recovery of heart function after hypothermic preservation of isolated rat heart was investigated,and the efforts was also done to find the possible protein biomarkers which could reflect the quality of donor heart after the preservation.Methods:Twenty-four adult Sprague-Dawley rats were divided into four groups and undergone hypothermic heart preservation.After baseline indices of cardiac function (coronary flow,heart rate,left ventricular developed pressure,max±dp/dt) were measured during perfusion using Langendorff isolated rat heart model.The hearts were arrested and preservated at 4℃for 6h in St.Thomas solution alone,St.Thomas solution supplemented with either FDP(5 mM)or SM(16 g/L) or both.After cold preservation,functional measurements were repeated,and water content of heart tissue was quantified.The activities of CK and LDH in preservation solution was assayed,and the proteins in it were also concentrated with DOC-TCA method. Concentrated proteins were undergone SDS-PAGE electrophoresis and silver staining. The software ImageJ was used to treat the image of SDS-PAGE electrophoresis gel.The molocular weights of the proteins which had a obvious different in amount among the samples were determined and the relevance between those proteins and recovery of heart function(±dp/dt.max,LVDP,HR,CF),heart water content and activities of LDH,CK was analyzed.Results:At reperfusion 20 min after cold preservation,In SM group and FDP group, the activities of CK were significant lower than that in control(P<0.05).In SM+FDP group,the recovery of HR were significant better than that in SM group (P<0.05).There was significant difference in recovery of heart function,water content of heart tissue and activities of LDH and CK between SM+FDP group and control(P<0.05).From SDS-PAGE gel,protein band a,d,e and f were obviously different in amount amongthe samples.The molecular weights of those proteins were 14.8 kDa,30.9 kDa,36.3 kDa,41.7 kDa respectively.Among them,protein a and d were positively correlated with activity of CK,protein d was positively correlated with activity of CK and LDH,protein e was negative correlated with recovery of LVDP and -dp/dtmax,protein f was negative correlated with recovery of LVDP,±dp/dt.max.Conclusion:Supplement of Fructose-1,6-diphosphate(FDP) and Salvia Miltiorrhiza extract(SM) to St.Thomas solution could improve the protective effects on isolated rat heart,which had a good synergistic effect.Protein a,d,e and f were associated with myocardial damage,which would be worthy of further study.
