Objective: To observe the combination effect of Total Isoflavones of Pueraria(TIP) and VitD on the immune function of ovariectomized rats,the rats model was established,in order to enforce immunologic intervention of post-menopause women to enhance immune function provide experiment theory .Methods: 81 female sprague-dawley(SD) rats of 3-month old were divided randomly into nine groups,according to 4×2 factorial design: model group, high dose TIP group, middle dose TIP group , low dose TIP group, VitD group, high dose combination group,middle dose combination group,low dose combination group,and Sham-operate group. Rats of the SHAM group only underwent the resection of a bit fat.The rest groups all underwent the resection of bilateral ovaries. Feeding 3 months to establish ovariectomized rats animal model. Ovariectomized rats animal model lavage different dose TIP and VitD 3 months.Then all the rats were killed.Blood was taken from the orbital veins of the rats to exmaine the RBC-C3b receptor chaplet and RBC-IC chaplet, FCM to analysis lymphocytes subgroup.Extracting thymus and cipher organic coefficient; asepsis extracting spleen ,prepare spleen cell suspension, Haemolysis spectropho-tography to evaluate the antibody forming cells population,MTT method to determin lymphocytes proliferation,LDH release method to determin NK cytoactive.Results:1.Except the low dose TIP, other groups the body weight of SD rats all lower than those in the control group , the degree of inhibition was related with the administrated dose of TIP(P<0.05);there was no interaction on inhibit body weight growth combined use TIP and VitD .2.Spleen coefficient of middle and high dose TIP and middle and high dose of combination group higher than those in the control group(P<0.05), there was no interaction combined use TIP and VitD; TIP and VitD group is inaction on thymus coefficient compared with those in the control group (P﹥0.05), there was no interaction combined use TIP and VitD.3.Lymphocytes proliferation and NK cytoactive of high dose of TIP and high dose combined groups higher than those in the control group( P< 0.05 ) , ;low, middle and high dose of TIP group and those of combined groups antibody forming cell population higher than those in the control group( P< 0.05 ), and display evident dose-effect relation; VitD is inaction to those, there was no interaction combined use TIP and VitD.4.RBC-C3b receptor chaplet of low and middle dose TIP and low and middle combined groups higher than those in the control group( P< 0.05), RBC-IC chaplet lower than those in the control group(P< 0.05); middle,high dose TIP and VitD can increase CD4+ of ovariectomized rats(P < 0.05), middle,high dose of TIP and VitD and each combination group CD8+ lower than those in the control group( P<0.05),CD4+/CD8+higher than those in the control group, and display evident dose-effect relation,there was no interaction combined use TIP and VitD.Conclusion:1.Fit dose TIP can enhance cell immunity and humored immune function,elevate nonspecific immunity of ovariectomized rats.2.VitD can adjust cellular immune function of ovariectomized rats; not display VitD enhance TIP immunological regulation to ovariectomized rats.
