Effect Of Yishen Capsule On The Expression Of TLR4 Of Renal Tissue In The Diabetic Nephropathy Rats

Objective:To investigate the effect of yishe capsule on the expression of TLR4,IL-6 and IL-8 of renal tissue in the diabetic nephropathy rats.Methords:Totally 36 healthy male wistar rats were divided randomly into three groups:normal control group(control group),DN control group(model group),yishe capsule treatment group,12 rats in each group.The rat models were induced by right nephrectomy and peritoneal injection of low-dosage streptozotocin.All the rats received daily gavage for 12 weeks.Including body weight,monitoring of blood glucose,the urinary protein in 24h and the level of serum creatinine,urea nitrogen were measured.The renal pathologyical changes were detected under light microscpoe.Expression levels of TLR4,IL-6,IL-8 in renal tissue of rats were detected by immunofluorescence microscope.Results:Modeling two weeks body weight decreased significantly in rats,and in the course of experimentation after the body weight remained decreased range,whereas the control rats in the experimental period,weight gain;Model of blood glucose in rats during the experiment has been in a stable,moderate high blood sugar condition,while the control group,blood glucose was maintained at 5mmol/L or so;4 weeks when the rats of model group significantly increased 24h urinary protein was significantly higher than that with time,the continuation of 24h urinary protein was progressive increase in trend;Model rats for 4 weeks when the blood Scr,BUN compared with the same period no significant change in the control rats,12 weeks Scr, BUN compared with the control group were significantly increased,In 12w,urinary protein quanitity in 24h,Scr and BUN of model group rats were highter distinctly than those of control group.After treatment of yishen capsule,urinary protein quanitity in 24h,Scr and BUN were reduced distinctly than those of control group.Under light microscope,4-week model group shows mild mesangial matrix,some with mild atrophy or tubular lumen expansion; 12 weeks model group increased mesangial matrix,mesangial area widened. Yishen capsule of rat kidney tissue treated mesangial matrix proliferation of reduced glomerular basement membrane become clear,no significant broadening.Glomerular sclerosis and tubular to reduce the extent of injury;Under immunofluorescence microscope,4 weeks,renal tissue of rats of model group TLR4,IL-6,IL-8 expression level of the beginning is higher than the control group,no significant difference.12 weeks,the expression of TLR4,IL-6 and IL-8 of renal tissue in the diabetic nephropathy rats were highter distinctly than those of control group;The expression of TLR4,IL-6,IL-8 of renal tissue in yishen capsule treatment group were lower distinctly than those of control group.Conclusion:The expression of TLR4,IL-6,IL-8 of renal tissue in the diabetic nephropathy rats were highter distinctly than those of control group.After treatment of yishen capsule,urinary protein quanitity in 24h,Scr,BUN were distinctly reduced.The expression of TLR4,IL-6,IL-8 of renal tissue in yishen capsule treatment group were distinctly descreased.The result suggests yishen capsule can effectively delay the progress of the diabetic nephropathy.Clinical treatment for the yishen capsule of diabetic nephropathy has laid a experimental basis.
