Effect Of Yishen Capsule On Wnt、Nf-κB Of Renal Interstitium In The Diabetic Nephropathy Rats

Objective To observe the effect of Yishen capsule on the expression of Wnt4、PGSK-3β、 NF-κB of renal interstitium in the diabetic nephropathy ratMethods The rat models were induced by peritoneal injection of streptozotocin(STZ).32Male Wistar rats were divided randomly into four groups:Normal group served as a control, DN group, Yishen capsule treatment group, losartan potassium treatment group,8rats in each group. Yishen capsule group received daily gavages of Yishen capsule. Normal group and DN group received daily gavages of distilled water. All the rats received daily gavages for12weeks. The urinary protein quantity in24h,the level of serum creatinine、urea nitro were measured after tewelve weeks treatment.The renal pathological changes were detected under light microscope.Expression of Wnt4、PGSK-3β、NF-κB protein was detected by immunohistochemical method. Expression of Wnt4、PGSK-3β、NF-κB mRNA was detected by FQ-PCR.Results The levels of urinary protein quantity in24h> serum creatinine and urea nitro were significantly increased in model rats compared to control rats. The levels of urinary protein quantity in24h、serum creatinine and urea nitro were decreased in Yishen capsule treatment group and losartan potassium treatment group rats compared to model rats. A significantly increase in the levels of Wnt4、PGSK-3β、NF-κB were observed in model rats when compared with control rats. The levels of Wnt4、PGSK-3β、NF-κB were decreased by Yishen capsule or losartan potassium. In model rats, a significantly increase were found in the levels of Wnt4、 NF-κB mRNA in kidney tissues when compared to control rats. The levels of Wnt4、NF-κB mRNA were decreased by Yishen capsule or losartan potassium.Conclusion The expression of Wnt4、PGSK-3β、NF-κB of renal interstitium in the diabetic nephropathy rats were higher distinctly. After treatment of Yishen capsule, the expression of Wnt4、PGSK-3β、NF-κB of renal interstitium were distinctly decreased. The result suggests Yishen capsule can effectively delay the progress of the diabetic nephropathy.
