Effect Of Yishen Capsule On The SOCS3, TLR4 And IL-6 Expression In Renal Tissue Of The Diabetic Nephropathy Rats

Objective: To investigate the effect of Yishen capsule on the SOCS3, TLR4 and IL-6 expression in renal tissue of the diabetic nephropathy rats.Methods: 40 Wistar male rats were divided randomly into four groups: normal control group (N), DN control group (DN), yishen capsule treatment group (DN+Y) and losartan potassium treatment group (DN+L), 10 rats in each group. The rat models were induced by uni-lateral nephrectomy and intraperitoneal injection of low-dosage streptozotocin (50mg/Kg). All the rats received daily gavages for 12 weeks. Body weight, the urinary protein quantity in 24h and the level of serum creatinine, urea nitrogen were measured in the progress. The renal pathological changes were detected by light microscopy. Expression levels of SOCS3, TLR4 and IL-6 in renal tissue of rats were detected by immunohistochemistry.Results: In 4 weeks, the 24h urinary protein of DN group rats significantly increased, and was higher than those of the N group rats. In 12 weeks, Scr and BUN of DN group were significantly increased than those of the N group rats. After yishen capsule treatment, urinary protein quantity in 24h, Scr and BUN reduced distinctly than those of DN group rats. The differences of urinary protein quantity in 24h, Scr, BUN between Yishen capsule treatment and losartan potassium treatment had no difference in statistics. Under light microscope, in 12 weeks, kidney of DN group rats obviously enlarged, mesangial matrix show diffuse hyperplasia, expansion, glomerular basement membrane wrinkle or irregular incrassation, glomerular sclerosis, interstitial inflammation. DN+Y group and DN+L group rats had decreased mesangial matrix, glomerular sclerosis had been reduced the extent of injury. Under immunohistochemistry, the SOCS3, TLR4 and IL-6 of renal tissue in N group had gleam expression, and began to express highly in DN group. The expression of SOCS3 in yishen capsule treatment group was higher than those of DN group, but TLR4, IL-6 were lower distinctly.Conclusion: The above results showed yishen capsule could delay the progress of the diabetic nephropathy effectively. Clinical treatment for the yishen capsule of diabetic nephropathy had laid an experimental basis.
