Study Of Yishen Capsule On SIRT6 And Podocin In Renal Tissues Of Diabetic Nephropathy Rats

Objective:To study the effect of yishen capsule on SIRT6 and podocin in the kidney tissue of rats with diabetic nephropathy.Methods:Forty SD healthy rats were randomly selected,and 30 rats were randomly selected to establish a diabetic nephropathy model by single injection of streptozotocin?STZ?after left nephrectomy.In the resveratrol group,each rat in the Yishen Capsule group was given 625 mg�kg^-1�d^-1,and in the resveratrol group,each rat was given 30 mg�kg^-1�d^-1,the normal group and diabetic nephropathy group were given the same amount of distilled water every day,and gavage was given for 8 weeks.The body weight,blood glucose,and 24-hour urine protein quantification were measured at the 0,4,and 8weekends.8 weekends to detect blood creatinine,urea nitrogen,total cholesterol and triglycerides;take rat kidney tissue,HE staining and Masson staining,light microscope examination of renal tissue pathological changes,immunohistochemical detection of SIRT6,podocin expression in kidney tissue Happening.Results:1)Compared with the normal group,HE staining of renal tissue in DN group showed partial glomerular hypertrophy,and a small number of glomeruli appeared mild sclerosis;mesangial cell proliferation was obvious,mesangial area was significantly widened,and inflammatory cell infiltration was visible renal tubular injury,partial renal tubular interstitial fibrosis;Masson staining showed vacuolar degeneration of some renal tubular epithelial cells,mild dilatation of renal tubules;thickening of arteriolar wall;blood glucose,24h urinary protein quantification,blood creatinine,urea nitrogen,total cholesterol and triglycerides were significantly increased?P<0.05?,and body weight was significantly reduced?P<0.05?.Immunohistochemical results in the DN group showed that SIRT6 study in renal tissue and podocin study in renal podocytes decreased?P<0.05?;2)Compared with the DN group,the kidney pathological changes of the yishen capsule group and the resveratrol group were alleviated,and the body weight was significantly increased?P<0.05?.The urinary protein quantification,serum creatinine,urea nitrogen,total cholesterol and triglyceride were significantly increased in 24 hours Decrease?P<0.05?.The immunohistochemical results of the yishen capsule group and the resveratrol group showed that the study of SIRT6 in renal tissue and the study of podocin in renal podocytes increased significantly?P<0.05?;there was no significant change in blood glucose?P>0.05?,but still higher than the normal group.Conclusion:SIRT6 has a certain role in the pathogenesis of diabetic nephropathy.Yishen capsule may improve the doctopoin injury of podocytes in DN model by up-regulating the expression of SIRT6 in renal tissue of DN model rats,and then delay the progress of diabetic kidney disease.
