The Study On Clinical Effect And Signal Transduction Related To The Impact Of Enzyme BTK With Busuishengxue Granule In Chronic Aplastic Anemia

Objective: To observe the clinical effect of chronic aplastic anemia (CAA) treated with Busuishengxue granule(BSSXG)and the difference in expression levels of the enzymes Btk before and after treatment. Revel the pathogenesis of CAA and the mechanisms of Busuishengxue granule. In order to Provide the therapeutic effect mechanism about BSSXG treating CAA.Method: 124 cases of CAA were divided into two groups at random, 61 patients of the treated group were treated with BSSXG and 63 patients of the control group were treated with Zaizhang Shengxue tablet( ZZSXT) .The treated group was divided into kidney-Yang deficiency type and Kidney-Yin deficiency type according to Traditional Chinese Medicine syndrome differentiation.We observed the clinical effect of these patients treated with BSSXG and ZZSXT. Meanwhile, the expression of the enzymes Btk of BMSC was detected by Western blot and Immunofluorescence technique before and after treatment.Result:1.The clinical therapeutic effect was superior in BSSXG group to ZZSXT( P<0.05). BSSXG could alleviate CAA patients anemia symptoms and increase the peripheral blood cells significantly. In BSSXG group, its curative effect was superior in Deficiency of Kidney-Yang type to that in Yin deficiency type( P<0.05).2 .The expression of Btk was lower than the normal controlled group before treatment , there are regulated respectively in different degree after treatment, and the Deficiency Kidney-Yang has better regulated degree than the Deficiency Kidney-Yin.Conclusion:1.The effect of BSSXG to CAA was superior to that of ZZSXT. 2 .The Abnomal expression of Btk serum can affect the bone marrow hematopoietic and is related closely to CAA. 3. BSSXG inhibit the apoptsis of bone marrow hematopoietic stem/ progenitor cells by regulationing Btk which mediate the signal transduction pathway.It also can mediate cell adhesion,migration,regulation cell proliferation,survival,and regulate the bone marrow hematopoietic microenvironment,increase the proliferation and differentiation of hematopoietic stem/ progenitor cells. 4.The clinical therapeutic effect was superior in BSSXG group to ZZSXT,and the regulated degree of the enzymes Deficiency of Kidney-Yin type were superior to Deficiency of Kidney-Yang type. It also further proves the theory that Yang asthenia is easily treated but Yin asthenia is hard to be cured.
