Dynamic counter-current extraction technology is a novel method for traditional Chinese medicine. It has the advantages of high-efficiency, decreasing energy consumption and shortening extraction time. This paper applied orthogonal design method to optimize the countercurrent extraction process of Longmu Zhuanggu granules. The detailed extraction is described and the extraction characteristics are summed up through combining with a contrastive analysis to common extraction. Besides, quality evalution was studied both from determination of active ingredients and fingerprint spectrum.The L9 (34) orthogonal design was used for studying the countercurrent extraction process, and defined the rate of dry paste, the content of astragaloside and polysaccharide as the main study indicators on the basis of literature and pre-trial, the optimized dynamic countercurrent extraction process were as follows:1:12 as the ratio of material to solvent, and 20min as extracting time in gradient level at 80℃. The optimized process is fesible for countercurrent extraction of Longmu Zhuanggu granules. Compared to traditional extraction method, this novel extraction technology is solvent-saving, energy-saving and high-extraction-efficiency, it is also a scientific method for traditional Chinese medicine, suitable for being used in industrial manufacturing.We tried to create a fingerprint spectrum of dynamic counter-current extract to control the extraction process and quality. Methods:ultrasonic waves extraction and then n-butanol extraction for preparation of test solution. chromatographic conditions were as follows:gradient elution with methanol-0.05% phosphoric acid; detection wavelength:267nm; flow rate:0.8ml/min; column temperature:30℃; injection volume:20μL; elution time:95min. Results:We created the HPLC fingerprint spectrum of the extract of dynamic countercurrent and it is feasible by the methodology. Conclusion:This HPLC fingerprint spectrum can be used to control the quality of dynamic counter-current extract.The content of astragaloside was determined by use of HPLC. Method: using evaporative light scattering detector, determine the content of astragaloside within dynamic counter-current extract. Results:There is a good linear relationship between peak area and the sample volume within the range of 104.8mμg~2620mμg. R=0.9995; The average recovery was 100.06%, RSD=1.85%; Conclusions:The method is simple, fast, reliable, accurate and can be used to determine the content of astragaloside of countercurrent extract.
