OBJECTIVE: To elucidate the expression of CD44 splice variant V6 (CD44V6), P-glycoprotein (P-gp) and Topoisomeraseâ…¡(Top-II) in CRC and their clinical significance.METHODS: Hematoxylin-Eosine method validation with colorectal cancer and normal mucos tissue. Immunohistochemistry was used to examine CD44V6, P-gp and Top-II expression in 80 colorectal carcinoma and 46 normal intestinal tissues. The relationship between CD44V6, P-gp and Top-II expression with sex, Dukes staging, tumor invasion and lymph nodes metastasis were statistically analyzed.RESULTS: The expression of CD44V6, P-gp, Top-II in CRC is higher than that in normal tissues. The expression of CD44V6 was positively related to the Dukes staging (rs=0.328,p<0.05), tumor invasion (rs=0.356,p<0.01) and lymph node metastasis (rs=0.406,p<0.01), but the sex was not . The expression of P-gp was not related to the sex, Dukes staging, tumor invasion and lymph node metastasis. The expression of Top-II was not related to the sex, Dukes staging, degree of invasion, but related to the degree of lymph node metastasis (rs=0.245,p<0.05).CONCLUSIONS: P-gp, Top-II and CD44V6 are highly expressed in CRC. CD44V6 was positively related to the Dukes staging ,tumor invasion and lymph node metastasis.
