Objective:①To establish chromatographic fingerprint of Nigella glandulifera Freyn et Sint and analyze it;②In order to find some better effective ingredients, We made a systematic research on Nigella glandulifera Freyn et Sint. Methods:①the gradient elution mode was applied in establishing HPLC fingerprint of Nigella glandulifera Freyn et Sint and comparing the 10 different origin of Nigella glandulifera. by "Evaluation Standard System to the similarity of fingerprints in Chinese traditional medicine (2004)".②The chemical constituents were analyzed by gas chromatography-mass spectrometry (GC-MS), which were extracted from Nigella glandulifera. through Supercritical carbon dioxide fluid extraction (SFE-CO2). The chemical investigation on the ethanol extracts of Nigella glandulifera was carried out by silica gel, Sephadex LH-20 and ODS RP-18 .Results:①Established the fingerprint of Nigella glandulifera. There were evident similar in them in despite of different growing environment.②Twenty-six compounds were identified by GC-MS, of which there are 16 volatile components and 10 fatty acids. The main components of fatty acid are telfairic acid (67.05%),oleic acid (19.15%), hexadecanoic acid(11.03%),the main volatile components are 9,12-octadecadienoic acid(68.58%),2-isopropyl-5-methyl-2,5-cyclohexadiene-1,4-dione(10.95%),22,23-dedihydro stigmasterol(6.93%),3,7,11-trimethyl-1,6,10-Cyclododecatriene-3-alcohol (3.43%),β-vitamin E (2.00%). Twelve compounds were isolated, and their structures were identified by physico-chemical characteristic and spectra data as 3-O-[D-xylopyranosyl -(1→3)-α-L-rhamnopyranosyl-(1→2)-α-L-arabinpyranosyl]-28-O-[-α-L-rhamnopyranosyl-(1→4)-β-D–glucopyranosyl-(1→6)-β-D-glucopyranosyl]hederagenin(1), 3-O-[D-xylopy ranosyl-(1→3)-α-L-rhamnopyranosyl-(1→2)-α-L-arabinpyranosyl]-28-O-[-β-D-glucopyr anosyl-(1→6)-β-D-glucopyranosyl]hederagenin(2), 3-O-[α-L-rhamnopyranosyl-(1→2)-α- L-arabinpyranosyl]-28-O-[β-D-glucopyranosyl-(1→6)-β-D-glucopyranosyl] hederagenin (3), Nigeglnaoside (4), Nigeglanine (5), 2-methyl-5-isopropyl pairphenolic (6), stigmasterol (7) and 1-O-hexadecanolenin (8). Compound (6) was isolated from this plant for the first time. Conclusion :①The fingerprint of Nigella glandulifera reflect the intrinsic quality completely and provide comprehensive methods of identification and quality evaluating for Nigella glandulifera. It is necessary for the modernization of traditional Chinese medicine.②This dissertation made a systematic research on Nigella glandulifera Freyn et Sint. as well as provides the basis for study of this plant.
