Purpose:(1) Association between Gln223Arg Polymorphism in Exon 6 of OB-R Gene and elite endurance athlete status.(2) Association between Gln223Arg Polymorphism in Exon 6 of OB-R Gene and the effects of endurance training.Subjects:â‘ Elite endurance athletes:They comprise 123 unrelated Chinese elite endurance athletes.They were specialists in long-distance running including 5000m,10000m and marathon.â‘¡Sedentary control:They comprise 206 unrelated Chinese sedentary undergraduates.â‘¢New soldiers: They consist of 102 unrelated new Chinese man soldiers.All subjects are Nan population and plainsmen in North China(including Liaoning,Heilongjiang,Jilin,Hebei,Tianjin and Shandong province).Methods:PCR-RFLP and Sequencing were used to examine the genotype and allele frequencies of Gln223Arg polymorphism in three subjects.The new soldiers undertook a distance running program consisted of 5000 m running with the intensity of 95-105%ventilatory threshold(VT),3 times/week for 18 weeks.VO2max,running economy(RE),body composition and Left ventricular structure and function were measured before and after the protocol,and were used to explore the association between Gln223Arg Polymorphism in Exon 6 of OB-R Gene and the effects of endurance training.Results:1.There were significant racial differences in the genotypes and allele frequencies in OB-R Gene Gln223Arg Polymorphism.2.There were significant differences in the genotypes and allele frequencies between males and females.3.There were significant differences in the genotypes and allele frequencies in OB-R Gene Gln223Arg Polymorphism between female EEA and SCON.4.There were no significant differences in the genotypes and allele frequencies in OB-R Gene Gln223Arg Polymorphism between male EEA and SCON.5.The PWS and reEF of A allele carriers(AA+AG) was higher than noncarriers(GG) before training.6.Theâ–³50W/SV of A allele carriers was significantly higher than noncarriers after training.Conclusions:The present study demonstrated that A allele of OB-R gene Gln223Arg polymorphism is associated with elite female endurance athlete status.There was slightly association between OB-R gene Gln223Arg polymorphism and training response.Heart fuction of A allele carriers may be more sensitive after training.
