Objective:To observe IL-6,GP130 and LIFR expression in rat myocardium before, during and after different periods of acute and long-term endurance exercise,identify in two different exercise circumstances whether IL-6, GP130 and LIFR have relativity or not.Methods:51 male SD rats were randomly divided into two groups, acute and long-term endurance exercise group.As the incremental load running with Bedford treadmill running model and treadmill running model of YunChang referenced,rats were killed accordance with points of design in experiment after last exercise.The rat myocardium was made into the tissue section for electron microscopy observation,HE dyeing and immunohistochemical staining. Capture the image by LEICA DM4000B,then calculate the percentage of positive cells.The date were analyzed by SPSS 13.0.Results:(1) After acute exercise, the morphostructure of the acute group and the endurance training group make a series of changes.(2) IL-6 In the acute group,the positive rate increases after acute exercise.It reach a peak at 1 hour after acute exercise, then gradually decreased.from 1 hour to 3 hours it increases obviously (P<0.01). In the endurance training group,the positive rate of endurance training group increases after acute exercise.It reach a peak at 3 hours after acute exercise (P<0.01), then gradually decreased. (3) GP130 In the acute group, the positive rate of GP130 increases extremely at 3 hours (P<0.01).There was no significant difference with the other groups.In the endurance training group, the positive rate of GP130 increases extremely at 3 hours (P<0.01). There was no significant difference with the other groups. (4) LIFR In the acute group, the positive rate increases after acute exercise.It reach a peak at 3 hours after acute exercise, then gradually decreased. The positive rate increases at 1 hour after acute exercise (P<0.05). It increases extremely at 3 hours and 5 hours after acute exercise (P<0.01). In the endurance training group, there was no significant difference with the other groups.Conclusion:(1) Acute exercise may cause the myocardial structure to damage mildly. The long-term endurance exercise causes the myocardium structure to have the compatible change. (2) After one-time exercise, the expression of IL-6, GP130 in the myocardium raised. Acute exercise can induce the myocardium in rats high expression of IL-6, GP130, and LIFR, which have a relationship with athletic heart. After endurance training, the peak of GP130 and LIFR is later than IL-6.That indicate that IL-6 can induce the expression of GP130 and LIFR. Description GP130 is likely to combine with the LIFR to form heterologous dimers,then transfer signal owing to the same peak time. (3) After long-term training in. rats, one-time exercise can induce high expression of IL-6, GP130 in the myocardium.But it is no significant change for LIFR. It is explained that rats had to adapt to the exercise load after endurance training. Endurance training affect on the different levels of different cell factors.
