| Dietary fiber has good physical and chemical characteristics, known as "theseventh nutrient", and it is widely used in food industry as nutritional fortifiers oradditives, which can improve the flavor and original structure of food and adjust avariety of physiological functions. Dietary fiber is composed of seaweed glue, semicellulose. As more hydrophilic groups in the structure of seaweed dietary fiber, it hasbetter water-holding capacity and it belongs to high soluble dietary fiber. Seaweeddietary fiber has better physiological functions which make it to be an ideal materialfor the development of healthy food.In this study, seaweed dietary fiber was compounded with the extracts fromastragalus, Pueraria lobata and folium mori according to the proportion of6.8:1.86:1:1.4and the hypoglycemic effect of the compound was studied. Diabeticmice model was prepared by tail vein injection of60mg/kg alloxan, then the modelmice were divided randomly into5groups: high-dose group, middle dose-group,low-dose group, model-control group and positive-control group. Blankcontrol-group was added additionally, which used orthoglycemic mice instead.Corresponding compound was administrated intragastrically, positive-control groupwas intragastrically administrated with200mg/kg metformin hydrochloride, modelcontrol-group and blank-control group used physiological saline instead.30dayslater, fasting blood glucose (FBG), triglyceride (TG), total cholesterol (TC), highdensity lipoprotein (HDL-C), low density lipoprotein (LDL-C) and oral glucosetolerance (OGTT) were tested. Mice pancrea was gathered by surgery for pathologyexamination.The results showed that FBG was extremely significantly decreased (p<0.01),the weight-loss was significantly alleviated (p<0.05) by high or middle-dose ofcompound and abnormal lipid metabolism was alleviated in a certain extent.Pathology examination revealed that the damage of β cells was effectively alleviatedby high or middle-dose of compound. Finally, INS-1cells were made to be the model of oxidative stress induced byalloxan, by which the antioxidant effects of extracts from Chinese traditionalmedicine were studied. The results showed that crude astragalus polysaccharide,total flavonoids from folium mori or Pueraria lobata can reduce oxidative stress inINS-1cells, alleviate the damage of alloxan to INS-1and promote the biosynthesisand secretion of insulin.The paper showed that compound of seaweed dietary fiber exhibitedhypoglycemic effects and provided a theoretic foundation and experimental proofsfor utilizing seaweed dietary fiber in hypoglycemic functional food. |
