| Traditional Chinese medicine is one of the most precious treasure of our Chinese nation, its compound health food has many advantages in prevention and control of diseases, such as its significant effect, non-toxic property and relatively cheap price, etc. With the development of the society, traditional Chinese medical health food gains more and more attention all over the world. As the saying goes,"Food recuperate is better than medical treatment", the essence of traditional Chinese medicine guides it to be suitable for a long-term safe health food of a new generation by reasonable compatibility, and it’s accepted by more and more people. Aloe is acclaimed by the Food and Agriculture Organization of the United Nations (FAO) as "One of the best healthy food products in the21st century". Hainan has appropriate climate and unique natural conditions, so Aloe barbadensis Miller is a kind of rich resource. Being as a traditional south medicine resource, it has a broad application prospect to make preparation of highly active and secure ingredients to develop a traditional Chinese medical compound health food of clear functional factors. The main work in the thesis is as follows:1. The aloe polysaccharide extraction technology was studied, response surface method optimization of four factors and three levels was processed by the Design-Expert software based on five single factor tests. And a high fitting degree, small experiment error and good repeatability mathematical model was established. Results showed that the optimum technique of ultrasonic-assisted extraction of polysaccharide from gel of Aloe barbadensis Miller was as follows:ultrasonic power was600W, ultrasonic time was45min, temperature was70℃, liquid-material ratio was5:1, pH was8.9and extraction time was once. In this condition, the percentage of extraction was0.1511%, which differed insignificantly with the predictive value.2. The deproteinization technology of polysaccharide from gel of Aloe barbadensis Miller was studied, the deproteinization rate of protein and the retention rate of polysaccharide were as comprehensive evaluation index, deproteinization effect of five methods including TCA, Sevag, papain, papain-TCA, papain-Sevag on polysaccharide were compared. Results showed that papain-TCA method was the optimum deproteinization technology and its operation was as follows:the amount of papain was2%, enzymatic time was1h, temperature was60℃, pH was6and followed by one treatment with5%TCA. Under this condition, the rate of deproteinization was83.4%, and the retention of polysaccharide was74.9%. The polysaccharide’s structure was analyzed by IR spectrum, UV spectrum,1H-NMR spectra and TG, comparison results showed that the characteristics of aloe polysaccharide were retained and some characteristic peak were reinforced to be easier to identify, the characteristic peaks of other impurities were eliminated or significantly weakened.3. The accelerated solvent extraction (ASE) technique of aloin from skin of Aloe barbadensis Miller was studied, optimization of four factors and three levels was processed based on three single factor tests. Results showed that the optimum technique of accelerated solvent extraction of aloin was as follows:extraction time was5min, extraction temperature was50℃, ethanol concentration was80%and extraction time was once. In this condition, the rate of extraction was1.91%. And the HPLC methodology was studied to determine its content.4. A kind of traditional Chinese medical compound health food granule was prepared by mixing aloe extract, Haematococcus Pluvialis. powder, Amomum villosum Lour, powder, Morinda citrifolia Linn, juice and other pharmaceutical excipients. Its quality standard was established, introducing the produce method, its traits and name, etc. TLC identification method was established by testing aloin, astaxanthin, bornyl acetate and scopoletin by refering to the2010version of "Chinese Pharmacopoeia". It was regulated that the granularity, moisture, ash content must not exceed15.0%,15.0%and7.0%respectively, and heavy metal such as lead, total arsenic and total mercury content must not exceed2.0mg/kg,1.0mg/kg and0.3mg/kg respectively. The content of functional components such as aloin, O-acetyl, astaxanthin, bornyl acetate and scopoletin shall be not less than4.00mg/g,3.90mg/g,1.20mg/g,2.40mg/g and170μg/g, and their methodology was studied, based on the pharmacopoeia and related standards.Finally, according to the ability of removing DPPH free radica and reducing power, the compound health care food preliminary showed a certain extent of antioxidant activity in vitro. |
