Gardenia jasminoides Ellis is a commonly-used traditional Chinese herbal medicine with a wide range of pharmacological activities.Gardenia yellow pigment and geniposide are the two main chemical components in the fruit of gardenia jasminoides Ellis.Gardenia yellow pigment,with good water-solubility,has been widely used as food additive and industry dye.At present,the production technology of crocin is complex with the low yield and the low colority.Usually the geniposide is general to be exhausted directly with the waste liquid,leading to the resources wasted.In order to develop useful techniques for producing of gardenia yellow pigment with high quality,we carried out the following works:1.Ethanol extraction,whisk-extraction and ultrasonic extraction used in the extraction of gardenia yellow pigment were discussed,and the relevant optimum conditions were determined with orthogonal experiments.The results showed that the whisk-extraction excelled the ultrasonic extraction and the traditional method in the yield at lower temperature and shorter time.The optimum conditions were determined as follow using ethanol/water(v:v=3:2) as the solvent,extracting 0.5 h,at 40℃,and the gardenia fruit/solvent ratio was 1:14. The yield of the extracted pigment achieved 98.1%,respectively.And we got the contents of the pigment in the used sample of gardenia jasminoides Ellis was determined as 6.20g/100g.2.In order to decrease the quantity of geniposide and increase the stability of gardenia yellow pigment,Macroporous adsorption resins were used to isolate and purify gardenia yellow pigment and geniposide.The adsorption characteristics of 11 kinds of resins for gardenia yellow pigment were investigated.By the experiments on the static unit volume adsorption capacity and the selectivity of every kind of resins for crocin,YWD09A5macroporous adsorption resin was chosen as the adsorbent of the crocin because it had good selectivity to crocin and could separate crocin and geniposide effectively,it was suitable to purify gardenia yellow pigment.The adsorptive quantity and adsorptive rate of crocin adsorbed by YWD09A5macroporous resin are the most.3.The separation result of the crocin with macroporous absorption resins column chromatography was studied.Regarding YWD09A5 macroporous adsorption resin as the research object,the experiments on the adsorption curve of the resin for crocin,effect of ethanol concentration on the behavior of the resin adsorption, the dynamic leaking curve under the different initial concentration,the dynamic leaking curve under the different flow rate were made and better conditions of YWD09A5 absorbing crocin were got.2BV/H as the flow rate on adsorption, 10mg/mL as the initial concentration,2.5ml/min as the flow rate on desorption and stepwise elution were adopted.Using these procedure,the color value (E1cm1%,440nm) and O.D value(A238/A440) of gardenia yellow pigment were obtained reaching more than 580and less than 0.2,respectively.4.An investigation of stability indicated that after adsorption and desorption for ten times,the performance of YWD09A5 resin comes to decrease.This paper mainly dealt with the extraction and purification conditions of gardenia yellow.The study laid theoretical foundation for large-scale extraction and purification of gardenia yellow.
