Gardenia fruit is rich in gardenia oil,gardenia yellow pigment and geniposide.In order to achieve high value utilization of gardenia fruit,this article used JinFu gardenia as raw material and used continuous phase change extraction to extract gardenia oil.A kinetic model for the extraction of gardenia oil by continuous phase change was established.The yellow pigment and geniposide were extracted by conventional solvent extraction and continuous phase change extraction.The LSA-10 resin was used to purify the gardenia yellow pigment.In the continuous phase change two-step extraction technique,the first step was to use 40%YH solution to extract geniposide,the second step was to use 75%ethanol solution to extract gardenia yellow,and the solvent and macroporous resin were used to refine the yellow pigment of gardenia.The extracts give yellow pigments of different color values.The main findings were as follows:?1?The optimal conditions for extraction of gardenia oil by continuous phase change were as follows:particle size 60 mesh,extraction time 100 min,extraction temperature 50°C,analytical temperature 60°C.Under this condition,the yield of gardenia oil was 16.21%.Eleven oils were identified by GC-MS analysis.The highest content was linoleic acid?48.023%?,followed by oleic acid?21.759%?,and the unsaturated fatty acid content was as high as 73.91%.?2?Based on the BaümLer model,a kinetic model for extracting sapilla oil by continuous phase change was established.The model fitted R2>0.95 to the experimental data,and the extraction temperatures were calculated to be 35,40,45,50,55 based on the fitted curve.At 60°C,the effective diffusion coefficients were 6.9341×10-7,8.750×10-7,9.7405×10-7,11.3925×10-7,9.9050×10-7 and 8.8322×10-7 m2/s,respectively.According to the fitted kinetic equation,the predicted extraction rate of the seed oil extracted at 35,40,45,50,55and 60°C for 50 min was predicted.By comparing the experimental results,the accuracy rate of the extraction rate was predicted by the kinetic model.Above 96%,the average accuracy is 97.75%.?3?The optimum extraction process of gardenia yellow pigment by conventional solvent extraction method was as follows:ethanol concentration 70%,extraction times 2,extraction temperature 60°C,single extraction time 60 min.Under these conditions,the extraction rate of gardenia yellow pigment was 93.65%,the geniposide extraction rate was 95.22%,the gardenia yellow pigment price was 60.17,and the OD value was 2.07.Continuous phase change extraction method selected 70%ethanol solution,extraction temperature60°C,analytical temperature 70°C,pump flow rate 40 L/h,extraction time was 2 h.Under this condition,the extraction rate of gardenia yellow pigment was 98.83%.The glycoside extraction was 98.01%,the gardenia yellow pigment price was 72.37,and the OD value was 2.02.?4?The adsorption and desorption rates of gardenia yellow pigment and geniposide by 7kinds of resins such as LX-30,LSA-10 and D101 were investigated,and LSA-10 resin was selected as the resin of refined gardenia yellow pigment.At a flow rate of 2 BV/h,the concentration of 3 mg/mL of gardenia solution was 2.5 BV,eluted first with 3 BV of tertiary water,and then 4BV 20%of ethanol was used to elute geniposide,followed by 4BV.The yellow pigment was eluted with an 80%ethanol solution to obtain a gardenia yellow product with a color value of 316 and an OD value of 0.42.The yield of gardenia yellow pigment was 89%.?5?The solubility of gardenia yellow pigment and geniposide in YH system was investigated.Finally,40%YH solution was selected as the solvent and continuous phase change extraction was performed for 6 h at 60°C.Under this condition,the extraction rate of gardenia yellow pigment was 29.32%,and the extraction rate of geniposide was 73.32%.In the second step,the crude extract of gardenia yellow pigment has a color price of 96.91and a OD value of 0.88.After extracting the geniposide crude solution with 0.5 times the mass of concentrated ethyl acetate,LSA-10 resin was used to adsorb gardenia yellow pigment,and it was determined that geniposide was eluted with 8 BV 20%ethanol solution,followed by 4 BV 80%.Ethanol solution was used to elute the gardenia yellow pigment to obtain a gardenia yellow pigment solution with a color value of 92.45 and an OD value of0.97.?6?Combine the two crude extracts of gardenia yellow pigment and purify them separately by solvent method and macroporous resin method.When the crude extract of gardenia yellow pigment was purified by solvent method,the gardenia yellow pigment extract was extracted and concentrated at 10 times 40%YH solution at room temperature.After extraction,the yellow pigment price of gardenia was increased from 96.91 to 165.34,and the OD value was increased.0.88 decreased to 0.512,the retention rate of gardenia yellow pigment was 96.99%,and the geniposide retention rate was 57.7%.When the macroporous resin method was used to purify the crude yellow pigment of gardenia,the geniposide was eluted with 5 BV 20%ethanol solution,and the yellow pigment was eluted with 3 BV 80%ethanol solution to determine the eluent A440 and solids.Content,calculated gardenia yellow color price as high as 738,OD value of 0.32.
