Gardenia is Gardenia plant ripe fruit, which is blindly commonly used Chinese herbal medicine, in most of our region has distribution. Gardenia Yellow element is one of the ingredients of major effective, mainly get from gardenia fruit extraction. Gardenia yellow element is a kind of natural water soluble yellow element, which is the kind of rare water-soluble carotenoids material and is often used as a natural food added pigment, Gardenia yellow element has some nutritional value, popular with pigment market. However, at present, the production product purity and quality can not satisfy for international standards.The experiment of the raw material is gardenia, it is lucubrate the condition which based on the traditional research of extraction, isolation, purification technology. Obtaining the high purity and pure gardenia yellow products, by the orthogonal test, the response surface method, macroporous resin column chromatography and polyamide column chromatography purification gardenia yellow element. The main research contents are as follows:1. Compare with the orthogonal experiment and the response surface method, and determined the best extraction process parameters. The orthogonal experiment for optimal results is60%ethanol, the ratio liquid and materials is1:10, Extraction temperature is70℃. After5hours, absorb light value is0.65. The response surface method for optimal results is58%ethanol. The ratio liquid and materials is1:10, extraction time is4.20hours, absorb light value is0.74experimental result is better than the orthogonal experiment. The best Extract Method is:58%ethanol, the ratio liquid and materials is1:10, extraction4.20hours.2. Organic solvent extraction preliminaryly purification coarse extraction liquid. Butarol is confirmed extracting agent. The specific value A238nm/A440nm is from3.15descend to2.48. After a series of post-processing get yellow powder which is Crude Product of gardenia yellow element. 3. Through the static-dynamic adsorption test, choosing D101macroporous resin to determine the Conditions is:sample flow velocity in0.50Bv/h, water and30%ethanol to remove impurities (geniposide).80%ethanol to remove gardenia yellow element. After a series of post-processing get gardenia yellow crude products. The specific value A23snm/A44onm is1.07. Colourity is192.70.4. Through the polyamide resin to get further purification. Determine the optimal conditions is:water to remove impurities (geniposide),20%ethanol to remove gardenia yellow element. After a series of post-processing get gardenia yellow products.
