| Object: To observe the influence of Jiangzhi Granule on the lipid metabolism and the vascular endothelial cell (VEC) function index of hyperlipemia model rabbits so as to investigate the intervention function and mechanism of Jiangzhi Granule on atherosclerosis (AS).Methods: 40 healthy rats were divided into 4 groups (10 rats every group), the blank group administrated with standard feedstuff, hyperlipemia model group administrated with hyperlipemia feedstuff, Jiangzhi Granule group administrated with hyperlipemia feedstuff and Jiangzhi Granule suspl. 2ml (2.7g/kg) intragastricly every day, Xuezhikang Group administrated with hyperlipemia feedstuff and Xuezhikang suspl. 2ml (0.625g/kg) intragastricly every day. After 4 weeks, what to be detected in the four group rats were the contents of serum lipid, NO, serum endothelin (ET), 6-keto-PGF1, and TXB2.Results: 1.Compared with the blank group, the level of TC, TG and LDL-C in the model group rats increased conspicuously (P<0.05 or P<0.01), and the level of HDL-C decreased conspicuously (P<0.01), which indicated it was successful to form the animal model. 2. Compared with the model group, the level of TC, TG, LDL-C, TXB2, and ET in Jiangzhi Granule group decreased conspicuously (P<0.05 or P<0.01), and the level of 6-keto-PGF1 and NO increased conspicuously (P<0.05 or P<0.01). 3. Artery morphology variation: diffuse stramineous plaques could be seen on the aorta inner wall of model group rats, especially more at the arch of aorta, distributed as irregular lamellar and protruding on the interface of inner wall. A few plaques could be seen only at the artery crotch of rats in Jiangzhi Granule group and Xuezhikang group, and distributed as diffusing mottling, which indicated light pathological changes. Through the observation with the light microscope in model control group, we could see the vessel wall thickened, not-smooth inner membrane, successive deficiency of VEC, underlayer collagen fiber revealed, a great deal mononuclear cells adhered to endothelium surface or dived into subendothelium, and subendothelial space widened. However in the Jiangzhi Granule group, the inner membrane was smoother, a few mononuclear cells adhered to the endothelium surface locally, it was not evident for VEC's proliferation, the deficiency of VEC could be seen, but better than the model group.Conclusions: Jiangzhi Granule can be used to treat hyperlipoidemia validly for it has the effect of regulating the balance of TXB2/6-ketoPGF1α and ET/NO to improve VEC, and relieving the damage of VEC in the earlier period of AS. |
