Herbal pair(HP) is the smallest unit of compatibility of traditional Chinese medicine.In this paper,three herbal pairs(HPs) were selected for the target of study.The chemical constituents of three HPs were investigated by GC-MS and HPLC-DAD technique combined with chemometrics analysis methods.These HPs include Herbra Ephedre(HE)-Zingiber officinale Rose(ZOR),Herbra Ephedre(HE)-Radix Puerariae (RP) and Cortex Phellodendri(CP)-Radix Glycyrrhizae(RG).The contents of the thesis mainly include the following sections:The first part(including chapter two and chapter three).The contents of the main constituents berberine and glycyrrhizic acid in CP-RG were analyzed by use of HPLC-DAD detection,and by comparing the chromatographic retention time and UV spectra of each pure component. At the same time,non-volatile components of the single herb and HP was compared qualitatively and quantitatively.In addition,also the effect of different ratios of the single herb on main ingredients were inspected.In chapter two,the changes of the chymistry composition were analysized in the decoctions of the single herb and HP.The effect of different ratios of the single herb on main ingredients were investigated. The conclusion was drew that the contents of berberine and glycyrrhizic acid in the decoctions of herbal pairs were lower than in the single herb.But the content of glycyrrhizic acid more significantly reduced in HPs.When ratio of CP and RG was 1:1,the content of glycyrrhizic acid become the lowest component in the content of glycyrrhizic acid of different ratios.Most components of CP-RG could be found in the HP which could be found in two sigle herbs.In chapter three,chemical components of precipitation which could dissolve in the simulated gastric fluid and intestinal fluid were compared qualitatively and quantitatively.The results showed that the sediments could partially dissolve in the simulated intestinal fluid and simulated gastric fluid.The composition is basically the same content between the simulated intestinal fluid and simulated gastric fluid.The content of composition is not very different.Its main components can be found in HP.The second part.Including the fourth chapter.the essential oils from single herbs HE,ZOR,RP and herbal pairs HE-ZOR,HE-RP were analyzed by gas chromatography-mass spectrometry(GC-MS).The pure chromatographic and pure spectra of each component were obtained by the use of heuristic evolving latent projections method(HELP).It can enhance the accuracy of qualitative and quantitative.The common compounds between single herbs and HPs were identified by the method of alternative moving windows factor analysis(AMWFA).The results showed that the herbal pair HE-ZOR has identified 81 chemical compounds,of which only 16 chemical compounds were from HE,only 43 chemical compounds were from ZOR.There are 8 chemical compounds among the three systems;15 new chemical components were only in HE-ZOR.For HP HE-RP which was identified 52 chemical compounds,in which 30 chemical compounds were from HE,and 17 new chemical components were found in HP,and there are 5 chemical compounds among the three systems.According to the numbers and content of the compounds in HE-ZOR and HE-RP,the dominant drugs are ZOR and HE respectively.
