Dryopteris fragrans (L.) Schott, a deciduous perennial herb in Dryopterideceae of Dryopteris Adanson, grows in sloping land with steatite where altitude is 1000-1700m and with stones of forest, or in the cracks of magma near the volcano. The Dryopteris fragrans (L.) Schott were often present in sub-community, which produce a lot of spores to make sexual reproduction and bed-out insemination. The plant distributes in Northeast and Huabei of China, Russia, Japan, Korea and North America.Dryopteris fragrans (L.) Schott has a strong physiologic activity and a wide range of civil applications. The inhabitant in northern Heilongjiang regarded the Dryopteris fragrans (L.) Schott as an unbeatable rival to skin diseases. According to the folk remedy of north Chinese, the Dryopteris fragrans (L.) Schott can treat all kinds of skin diseases and arthritis, such as psoriasis, rash, dermatitis, athlete's foot, dry skin and etc; they have obvious curative effects for psoriasis and arthritis. In recent years studies have shown that Dryopteris fragrans (L.)Schott has many effects such as anti-immune activity, anti-tumor and inhibit fungi, the treatment of psoriasis and so on.Although the Dryopteris fragrans (L.) schott with many anti-inflammatory effects, but its effective anti-inflammatory site has not been confirmed. In order to determine the effect parts for the treatment of inflammatory and to make sure further the active intergradient within the effect parts, Dryopteris fragrans (L.) Schott water extracts was eluted with water and ethyl alcohol in three different concentrations (50%, 95%) in macroporous resin. Then the four eluates were screened and studied as the active fraction for the treatment of inflammatory. It indicated that 50% alcohol elute of Dryopteris fragrans (L.) Schott was decided as major active fraction for the anti-inflammatory and its chemical ingredients were isolated. In this paper, fifteen compounds were isolated with silica. Gel column, ODS column, HPLC preparation chromatography and others modern extraction and separation technologies from effective parts of treating inflammatory. Furthermore, chemical structures of eight compounds were identified by 1D and 2D NMR such as 1H-NMR, 13C-NMR, HSQC and HMBC, combined with ESI-MS and chemical methods. Compound 8 was discovered for the first time in Dryopteris fragrans (L.) Schott .In order to further study the effective parts of the physiologic activity of compounds, the study on immunoregulation activity of some of the compounds was processed. Through the vitro lymphocyte transformation experiment, three compounds were determined with immunoregulation activity. In this paper, the pharmacological action of active fraction of the plant Dryopteris fragrans (L.) Schott for inflammatory treatment and effect substances were further clarified, which deliver the study and research of Dryopteris fragrans (L.) Schott used for anti-inflammatory medicine with important references. In a word, this research will have inspiring prospects and significant application. This program is supported by Heilongjiang Province Natural Science Fund.
