| Dryopteris fragrans(L.) schott, a deciduous perennial herb in dryopteris genus of dryopteris family, grows in 1000-1700m altitude of the sloping land with stones of high cold areas, the cracks of magma near the volcanoes and in the gravel slopes of forest. According to the Chinese folk remedies, the Dryopteris fragrans (L.) schott can treat all kinds of skin diseases and have obvious curative effects for athlete's foot and dry skin caused by the fungus.In recent years, the Dryopteris fragrans(L.) schott was studied on treatment of psoriasis and on anti-fungi, but the studies on dryopteris fragrans(L.) schott active intergradients were nearly reported in China. In order to determine the anti-fungi effect parts, the water extracts of Dryopteris fragrans(L.) schott were further extracted with petroleum ethers of different polarity, chloroform and N-butanol respectively.Then the N-butanol extracts were screened through AB-8 macroporous resin column and eluted by ethyl alcohol in three different concentrations (45%,75%,95%). Then the four eluates were screened and studied as the anti-fungi active fractions. Finally 45% alcohol eluate of Dryopteris fragrans(L.) schott were decided as the major active fraction for anti-fungi.In order to determine the chemical compositions within the effective components, 13 compounds were isolated from the 45% alcohol elutate of Dryopteris fragrans(L.) schott through silica gel column, ODS column, Sephadex column and HPLC chromatography, and other extraction methods. Furthermore, chemical structures of 6 compounds were identified by physical and chemical methods, and modern spectroscopy testing technologies such as 1H-NMR,13C-NMR,DEPT,HSQC, HMBC and ESI-MS mass spectrometry, including 2 sesqutierpenoids,1 lignins,2 flavonoids,1 aromatic series. Referring to literatures, a compound has been comfirmed as a new compound, which is 2-ethyl-5,7-dihydroxy-7-O-β-D-glucopyranosyl Chromone. Among five known compounds,3',4',5,7-tetrahydroxy-dihydrogen-flavonoid-7-O-β-D-glucopyranoside, trans-S-trans-caffeic acid-9-β-D-xylpyranosyl (1→2)-β-D-glucopy ranosyl ester,3-oxo-7,8-dihydrogen-α-ionol-9-O-β-D-glucopyranoside from were also the first time isolated from Dryopteris fragrans(L.) schott. The other known compounds were (6S,9R)-3-oxo-a-ionol-9-O-β-D-glucopyranoside and (7S, 8R)-dihydrodehydrod iconiferyl-9-O-β-D-glucoside.In this thesis, the anti-fungi active ingredients of Dryopteris fragrans(L.) schott were studied, the results will provide some important experimental data for further research on treatment of tinea caused by fungus, and will have practical significance for searching natural antimicrobial substances, meanwhile will play a positive role in promoting development of new drugs. |
