Dryopteris fragrans (L.) schott, a deciduous perennial herb in dryopteris family of dryopteris genus, grows in the cracks of magma near the volcanoes, and chiefly distributes in Wu-da-lian-chi of Heilongjiang province. According to the local chronicles, for hundreds of years, water extracts of D. fragrans (L.) schott have been used to treat many kinds of skin diseases, especially have obvious curative effects for psoriasis. The inhabitant in northern Heilongjiang regard D. fragrans (L.) schott as an unbeatable rival to skin diseases.As a traditional Chinese medicine, D. fragrans (L.) schott was reported on the treatment of psoriasis and fungi these years. However, the system study on D. fragrans (L.) schott's active ingredients and pharmacology for treatment of psoriasis was neither reported in China nor in abroad. In order to determine the effective component for the treatment of psoriasis and confirm the active ingredients in the effective component, the dry power of D. fragrans (L.) schott was eluted with water firstly,with ethyl alcohol secondly,with water thirdly. D. fragrans (L.) schott water extracts was concentrated into ointment in the low-pressure. Then the liquid of ointment was extracted by petroleum ether, chloroform and N-butanol. The extraction of N-butanol was separated with water and ethyl alcohol in three different concentrations (30%, 60%, 95%) in AB-8 macroporous resin column. Then the four eluates were studied of screening as the effective component for the treatment of psoriasis. At last, 30% alcohol elutate of D. fragrans (L.) schott was decided as major active fraction for the psoriasis.According to the studies on effective parts of treating psoriasis, 11 chemicals were isolated through silica Gel column, ODS column, HPLC chromatography. Furthermore, chemical structures of 5 chemicals were identified by chromatography and chemical methods.They were Rutin,3,5-dimethy-6-hydroxy-2-methoxy-4-O-β-D-glucopyranosyl phenyl hypnone, (1R,3S)-1-methyl-2,3,4,9-tetrahydro-1H-pyrido[3,4-b]indole-3-carbo- xylic acid, trans-p-hydroxy cinnamic methyl ester, quercetin-3-O-β-D-glucopyra- noside. Among them, Rutin, (1R,3S)-1-methyl-2,3,4,9-tetrahydro-1H-pyrido[3,4-b]indole-3- carboxylic acid and quercetin-3-O-β-D-glucopyra-noside were discovered for the first time in Dryopteris.All these research will provide important scientific foundation and material base for further research about psoriasis. The article provides significant scientific evidence for exploiting and utilizing the resource of the Dryopteris fragrans (L.) schott.
