Objective:Ginseng proteins and polypeptide, which have a variety of biological activities but less reasearches, were studied by modern technologies including isolation and purification technology, activity detection technology and analysis - identification technology. Providing a basis for identification and quality evaluation among ginseng, wild ginseng and American ginseng by showing a comparative study of their water-soluble protein components.Methods: Using different buffer, different ratio of solution, different extraction time to extract the ginseng protein, using Bradford method, determin the rate of protein extraction for the evaluation Standards, choose the best optimized extraction process. Purification was carried by hollow fiber system and column chromatography technology, combined with cell activity to screening activity component. Compare the difference of the proteins among ginseng, wild ginseng and American ginseng by using SDS-polyacrylamide gel electrophoresis (SDS-PAGE) and image analysis software.Results: Orthogonal experimental results show that the pH of buffer, and extraction time were all have an affect to the extraction rate of the total protein. The highest is pH7, liquid ratio of 1:15, extraction time of 18h. After hollow fiber membrane filtration system, the yield up to 3%, the purity up to 80%. a molecular weight of 5027Da peptides was separated and purificated from the total protein of ginseng, activity experiments show that it have inhibitory effect on 3T3 cells proliferation and have inhibitory effect on Hep-2 cells proliferation, Moreover, it has a certain dose-effect relationship. Electrophoresis results showed that the main protein of ginseng, wild ginseng and American ginseng were distribution in the 60.20-5.03kDa, however, the content are quite different. Ginseng's is hightest and wild ginseng's is lowest, there are only two higher content protein, with molecular weight of 60.20kDa, 16.50kDa,Several other protein content were all less than ginseng. American ginseng has three high content protein, with molecular weight of 60.20kDa, 25.20kDa, and 18.20kDa.Conclusion: The best extraction process of ginseng total protein is: neutral buffer solution extraction, liquid ratio of 1:15, extraction time of 18h. The use of membrane separation technology will make a high purity and yield of the total ginseng protein, it solved the problems of complicated operation, long cycle and higher costs during traditional process and it is suitable for industrial production. with the promotion of the role towards 3T3 cell but the inhibition of Hep-2 cell, it shows that its has promotion affect of fibroblast growth and inhibition affect of cancer cell growth. It has provided a new method to identificate and evaluate the quality of ginseng, wild ginseng and American ginseng.
