Objective: Observe nonalcoholic fatty liver disease(NAFLD) rat skeletal muscle cell autophagy, the changes of aerobic exercise on NAFLD rat skeletal muscle autophagy related protein expression level of influence.Methods: 50 SD rats were randomly divided into 5 groups: 8 weeks, 16 weeks blank control group(C8, C16), average feed;8 weeks, 16 weeks NAFLD model group(M8, M16 group), exercise intervention group(EM), high fat feed.8 weeks after testing C8 and M8 group of liver and blood biochemical indicators, to determine the incidence of NAFLD.Since then, the EM group 60 min/d, 6 d/w eight weeks without weight loading swimming movement, observe C16, M16 and EM group of liver cells form and HE staining;Determination of blood glucose(FPG), insulin(FINS), aspertate aminotransferase(AST), alanine aminotransferase(ALT), total cholesterol(TC), triglyceride(TG) value, calculate the indexes of insulin resistance(HOMA IR);Electron microscope observation of gastrocnemius ultrastructure, Western blotting test skeletal muscle autophagy protein LC3, Beclin-1 protein expression.Results: 1. HE staining C8 group of liver cells did not see obvious steatosis, M8 group has 8 of 10 only apparent hepatocyte steatosis, cell vacuoles, balloons, disordered arrangement of liver cells, did not see necrotic lesions, portal area without inflammatory cell infiltration and fibrous tissue hyperplasia;C16 group of only 2 only a small hepatocellular fatty degeneration, M16 group and 10 were obvious hepatocyte steatosis, portal area with hepatic sinus expansion, blisters, hepatic lobule inflammation;EM group 9, only 6 only visible liver steatosis, degree of the M16 group light, no inflammation, necrosis or fibrous tissue hyperplasia.2. Compared with C8 group, M8 group of serum ALT, AST, TC, TG content were increased, and the TC value is statistically significant(P <0.05), and the rest did not see significant difference(P > 0.05);Compared with C16 group, M16 group of serum ALT, AST, TC, TG content were increased, but no significant difference(P >0.05);EM the M16 group serum ALT, AST, TC, TG content reduce, levels, but did not return to C16 group were no significant difference(P >0.05).3. Compared with C16 group, higher M16 group of FPG, FINS, HOMA- IR index increased, with statistical significance(P < 0.05);LC3, Beclin1 autophagy protein decreased, has the extremely significant difference(P < 0.01).4. Compared with M16 group, EM group of FPG, FINS and HOMA- IR index is reduced, no significant difference(P > 0.05);And did not restore to C16 group level, with statistical significance(P < 0.05);Compared with M16 group, EM LC3, Beclin-1 autophagy protein expression levels rise, no significant difference(P > 0.05);And failed to reach C16 group level, has the extremely significant difference(P < 0.01).Conclusion: 1.8 weeks a high-fat diet can induce 80% NAFLD SD rats, over time, the fat of NAFLD incidence degree and aggravating.2.NAFLD rat skeletal muscle cell autophagy is reduced, show the LC3- II, Beclin-1 protein expression of the ratio of volume LC3- II/LC3- I cut, 8 weeks swimming exercise can make the above index rise, prompt, enhance the level of skeletal muscle autophagy aerobic exercise can be have certain protective effect on skeletal muscle cells.
