Objective: Observation movement on the AngⅡamount of plasma and Aorta,the NO amount and RhoAmRNA,ROCKmRNA expression of the Aorta of hypertensive rats,and to explore appropriate exercise on the prevention of complications of hypertension and effect and mechanism for the prevention and treatment of hypertension and its complications and provide a theoretical basis.Methods: 16 male Wistar rats were divided randomly into control group(Group WC)and training(Group WT).16 male SHR rats were divided randomly into control group (Group SC)and training(Group ST), with 8 in each. Group WT and Group ST daily 60-minutes swim without load movement for 6 days per week during the continuous 8 weeks. During the test period, the blood pressure and weight of rats were tested every week.8 weeks later, we respectively measured the NO amount of the Aorta, the AngⅡamount of plasma and Aorta, the activity of NOS,iNOS of the Aorta, and the expression of RhoAmRNA,ROCKmRNA,ROCK-1mRNA,ROCK-2mRNA of the Aorta in every single group.Results:1.After the 60 minutes swimming every day for 8 weeks, we found the weight in Wistar training Group was obviously lower than control Group(P<0.01), and the weights in SHR training Group was obviously lower than control Group(P<0.01).The blood pressure in SHR training Group obviously descended more than that in control Group (P<0.01). It means that sports can control blood pressure by reducing body weight in SHR rats.2. After the 60 minutes swimming every day for 8 weeks, the AngⅡamount of myocar in SHR training Group and control Group significantly rose compared with that in Wistar control Group(P<0.05), Wistar training Group rose than control Group, but no conspicuous difference(P>0.05), and SHR training Group lower than control Group, but no conspicuous difference, either(P>0.05).The AngⅡamount of the Aorta in SHR training Group conspicuously declined compared with that in control Group(P<0.05), the AngⅡamount of the Aorta in Wistar training Group conspicuously declined compared with that in control Group(P<0.05), too. It means that sports is on plasma AngⅡn o significant effect, but reduce the content of AngⅡon rat aorta.3. After the 60 minutes swimming every day for 8 weeks, the NO amount,NOS activity of the Aorta in SHR training Group conspicuously rose compared with that in control Group(P<0.05),and the NO amount,NOS activity of the Aorta in Wistar training Group conspicuously rose compared with that in control Group(P<0.05),too.The iNOS activity of the Aorta in Wistar training Group compared with control Group rose, but no big difference(P>0.05),iNOS activity of the Aorta in SHR training Group compared with control Group rose, but no big difference(P>0.05).It means that sports promote the synthesis of NO release in rats, increased NOS activity, reduced blood pressure.4. After the 60 minutes swimming every day for 8 weeks, the expression of ROCKmRNA of the Aorta in Wistar training Group conspicuously ascended compared with control Group(P<0.05),but the the expression of RhoAmRNA compared with control Group ascended, but no big difference(P>0.05).The expression of RhoAmRNA,ROCKmRNA,ROCK-1mRNA of the Aorta in Group SHR training Group conspicuously ascended compared with control Group(P<0.05), but the the expression of ROCK-2mRNA compared with control Group ascended, but no big difference(P>0.05). It means that sports down-regulating RhoA / ROCK pathway activity to achieve step-down purpose, ROCK-1 sub-type plays an important role. Conclusion:1. Long-term appropriate and regular exercise can significantly inhibit the further increase of SHR blood pressure, and can mitigate the hypertension. Meanwhile in works on inhibiting the weight of Wistar and SHR rats from increasing.2. Long-term appropriate and regular exercise can decrease the AngⅡamount of the Aorta of SHR and Wistar, reduce the pressor effect of AngⅡ, which plays quite an active role of preventing and reducing high blood pressure.3. Long-term appropriate and regular exercise can increase NO amount of the Aorta of SHR and Wistar ,and improve NOS activity.It is indicated that exercise can promote the release of NO synthesis, enhanced NOS activity, so that recovered vasodilatation, decreased blood pressure.4. Long-term appropriate and regular exercise can significantly inhibited SHR rat aortic RhoA / ROCK pathway by a number of ways,for example, by reducing the content of AngⅡin turn prevent more activated RhoA and ROCK, increases NO and enhancing the activity of NOS inhibition of RhoA / ROCK pathways of activity, finally decreased blood pressure.
