Prunus maritime (Prunus, Rosaceae), a deciduous shrub, originated from the northeast America, which is widely used in the gardens and ecological protection owing to its wide adoption and tolerance to stress, numerous large flower, and colorful and good taste fruits.In present study, the germplasm introduction, propagation and container seedling were investigated. Prunus maritima was introduced into Wujiang, Jiansu province from America. Seeds were stored at different conditions and treated with different seed soaking methods, optimal conditions for germination and seedling were determined. Micropropagation was also studied, one-year old new shoots was employed as explants, MS was used as basic medium. Effects of plant growth regulators such as auxins (NAA, IBA and 2,4-D) and cytokinins (6-BA, KT) on the adventitious bud multiplication and rooting were studied. In addition, technology for container seedling was studied too.The results are as follows:1. Introduced Prunus maritima performed well at Wujiang area, it seems that it is a good shrub deserving popularity for its high ornamental, economic and ecological values.2. Wet sand storage stratification at temperature no more than 3℃and cold water seed soaking pretreatment can accelerate seed germination and enhanced the seed emerging percentage as high as 69.7%.Otherwise, worse germination performance was observed in seeds without cold stratification.3. Seeds with thousand seed weight>270g are matured and full, and gave higher rate of seedling emergency, whereas, seeds with thousand seed weight<270g gave lower, exhibited lower seed germination.4. One-year old shoot was used as explants, MS medium supplemented with 6-BA (0.1mg/L) and NAA (0.05mg/L) was optimal to bud break, Multiplication index, as high as 10, of adventitious bud was obtained on MS+6-BA 0.5 mg/L+2,4-D 0.1 mg/L.93.7% of adventitious shoots rooted on 1/2MS+6-BA 0.5 mg/L+NAA 0.2 mg/L after being cultured for 20 days,4.5 roots per shoot on average was observed.5. Cultivation medium for container seedling, peat soil is good for height, while for growth, bark is better than perlite and sawdust. Application of'Aiyinshi'slow release fertilizer 2-4Kg/m3 is appropriate. Peat soil:perlite (1:1) was suitable for transplanting of regenerated seedling, and gave higher survival rate of seedlings.
