Objective:To establish sciatic nerve injury model in rats, to observe the effects of Erdunwurle on early c-fos expression and neurological function after the sciatic nerve injury, to explore the efficacy and mechanism of the drug on sciatic nerve injury.Methods:Total of80SD rats(170-230g weight)were randomly divided into five groups:model group, blank control group, positive control group, high doses group of Erdunwurle, low doses group of Erdunwurle, each group was16rats. In addition to the blank group anaesthetize with10%chloral hydrate intraperitoneal injected before exposed the sciatic nerve, and crushed sciatic nerve with haemostatic to transparent membrane before closed the incision except that the sciatic nerve of blank control group rats was exposed before close the incision but not crushed sciatic nerve. Low doses group of Erdunwurle rats and high doses group of Erdunwurle rats was gastric lavaged suspension of Erdunwurle0.3g/kg and1.2g/kg, positive control group rats give the vitamin B1(3g/kg)+vitamin B12(10g/kg), the volume of gastric lavaged was2ml/100g for4weeks continuously, twice a day. The model group rats and the blank group rats were gastric lavaged with distilled water of same volume. Each group rats choose8rats observed the SFI, TWL and NCV at1st and4th week postoperative. Sciatic nerves and L4-6segment of spinal cords were dissected for pathological changes observation and c-fos expression in spinal dosal horn in the immunohistochemical staining method. The results were do F-test.Results:(1) SFI:At1st week, SFI of low doses of group Erdunwurle were significantly higher than the model group (p<0.05).(2) TWL:Compared with the model group, low doses group of Erdunwurle was apparently prolong postoperativefor1st week (p<0.05).(3) NCV:At1st week, NCV of the model group were slower than the NCV of the others (p<0.01). At4th week, NCV of low doses group of Erdunwurle were significantly faster than the model group (p<0.05).(4) The gray value of c-fos expression: At1st week, the gray value c-fos expression of model groups were significantly lower than the others(p<0.05), in the low doses group rats of Erdunwurle, the gray value c-fos expression were significantly higher than that in the high doses group rats of Erdunwurle and positive control group rats (p<0.05). At4th week, in the low doses group rats of Erdunwurle, the gray value c-fos expression were significantly higher than that in the model group rats and high doses group rats of Erdunwurle (p<0.01), in the low doses group rats of Erdunwurle, the gray value c-fos expression were significantly higher than that in the positive control group rats (p<0.05).Conclusion:(1) Erdunwurle has obviously promotion effects for recovery of nerve functional of sciatic nerve injury in rats.(2) Erdunwurle has analgesic effects on the sciatic nerve injury in rats.(3) Erdunwurle is capable of promoting the nerve conduction velocity of sciatic nerve injury in rats.(4) Erdunwurle has inhibited c-fos expression role after the sciatic nerve injury.
