Objective:This topic by observing the clip ridge electric acupuncture combined with neural mobilization to rabbit sciatic nerve conformity of ethology score after injury,nerve conduction velocity detection,and cdc42 mRNA and protein expression,the influence of the clip ridge electric acupuncture combined with neural mobilization to hemi-resected lumbar rabbit sciatic nerve regulation mechanism,thus for electric acupuncture combined with neural mobilization provides the theory basis for treatment of sciatic nerve injury.Methods:Will be 180 New Zealand big-eared rabbits using clamps the preparation of rabbit sciatic nerve Sunderland ? damage conformity model,were randomly divided into 5 groups:normal control group,model control group,nerve manipulation group,the clip ridge electric acupuncture group and electric acupuncture combined with nerve manipulation group(combination group).Each group of 36 patients was divided into 3 subgroups at 1 week,2 weeks and 4 weeks after surgery,with 12 in each subgroup.The sciatic nerve injury model was caused by clamping method.Normal control group,model control group without any intervention,manipulation group line nerve for the treatment of loose,clip to clip ridge ridge electric acupuncture group cupping treatment,clip ridge electric acupuncture combined with nerve manipulation group including clip ridge and nerve manipulation treatment.After 1 week,2 weeks and 4 weeks after treatment,the materials were taken for behavioral assessment,nerve conduction velocity detection,and the expression of cdc42 gene and protein in the sciatic nerve and spinal cord were detected by WB and PCR.Results:1.The toe reflexes and improved Tarlov scores three time points score were higher than that of model control group,treatment group is lower than the normal control group(P<0.05),and combining with the above clip ridge electric acupuncture group and nerve manipulation group(P<0.05),of which 4 w point in time restore best;2.Compared with normal group,the nerve conduction velocity of the other groups decreased,and the difference was statistically significant(P<0.05).In addition,the combination group was higher than the clipping group and the nerve loosening group(P<0.05),and the nerve conduction velocity was the highest in 4W.3 Sciatic nerve(1)The expression of Cdc42 protein was detected by WB:compared with the normal group,the expression of Cdc42 protein in the remaining groups decreased,and the difference was statistically significant(P<0.05).Compared with the model group,the expression of Cdc42 protein was increased with the increase of Cdc42 protein expression(P<0.05).Moreover,the expression of the clipped electroacupuncture combined with the nerve loosening group was significantly higher than that of the single treatment group,and the difference was statistically significant(P<0.05).(2)PCR detection of Cdc42 gene expression:compared with the normal group,the expression of Cdc42 gene in the remaining groups decreased,and the difference was statistically significant(P<0.05).Compared with the model group,the expression of Cdc42 gene was increased with the increase of the expression of Cdc42,and the difference was significantly higher than that of the nerve loosening group,the clipping group and the clipping electroacupuncture group.Moreover,the expression of the clipped electroacupuncture combined with the nerve loosening group was significantly higher than that of the single treatment group,and the difference was statistically significant(P<0.05).4.Corresponding spinal segments.(1)The expression level of Cdc42 protein was detected by WB:compared with the normal group,the expression of Cdc42 protein in the remaining groups decreased,and the difference was statistically significant(P<0.05).Compared with the model group,the expression of Cdc42 protein was increased with the increase of Cdc42 protein expression(P<0.05).Moreover,the expression of the clipped electroacupuncture combined with the nerve loosening group was significantly higher than that of the single treatment group,and the difference was statistically significant(P<0.05).(2)PCR detection of Cdc42 gene expression level:compared with the normal group,the expression of Cdc42 gene in the remaining groups decreased,and the difference was statistically significant(P<0.05).Compared with the model group,the expression of Cdc42 gene was increased with the increase of the expression of Cdc42,and the difference was significantly higher than that of the nerve loosening group,the clipping group and the clipping electroacupuncture group.Moreover,the expression of the clipped electroacupuncture combined with the nerve loosening group was significantly higher than that of the single treatment group,and the difference was statistically significant(P<0.05).Conclusion:1.Neural mobilization,including ridge,clip ridge electric acupuncture combined with neural mobilization can improve behavior score after the rabbit sciatic nerve injury,improve nerve conduction velocity,promote the sciatic nerve and spinal cord segmental Cdc42 gene and protein expression,and clip ridge electric acupuncture combined with neural mobilization is superior to the pure nerve manipulation and simple clip ridge cupping treatment.2.The combination of spinal cord and nerve loosening may facilitate the expression of Cdc42 gene and protein,regulate the rearrangement of cytokines skeleton,regulate protein kinase,and promote axonal regeneration.
