Three experiments were conducted to assess the standardized ileal digestibilities (SID) of aminoacid in46corns, and to establish the near infrared reflectance spectroscopy (NIRS) calibration equation,in order to predict the standardized ileal digestible amino acid (AA) of corns for broilers rapidly byNIRS.Experiment1Study on the estimation of the basal endogenous amino acid loss for broilersThis experiment was conducted to determine the basal endogenous amino acid loss (BAAL) using theNitrogen-free diet.4828-day-old healthly Arbor Acres broilers were selected, and they were dividedinto eight repeats (4repeats for male and female respectively) randomly, each repeat had six chickens.Ad libitum to feed and water, chickens were sacrificed using electrical shock at the end of the trial. Ilealdigesta, collected form four chickens, were used to determine the BAAL at the terminal ileal of35-day-old broiler. The results showed that, the sex of broiler has no significant effect on BAAL(P>0.05). The determined BAAL of broilers were as follows: Asp327.68mg/kg DMI, Thr314.60mg/kgDMI, Ser288.96mg/kg DMI, Glu438.85mg/kg DMI, Gly191.61mg/kg DMI, Ala224.62mg/kg DMI,Cys135.06mg/kg DMI, Val194.06mg/kg DMI, Met47.75mg/kg DMI, Ile134.29mg/kg DMI, Leu262.97mg/kg DMI, Tyr218.67mg/kg DMI, Phe196.98mg/kg DMI, Lys147.05mg/kg DMI, Arg260.05mg/kg DMI, Pro225.04mg/kg DMI.Experiment2Determine of standardized ileal digestible amino acids in corns for broilers46corns were collected from different places, and semi-pure diets were formulated respectively, while thecorn as the sole nitrogen resources.220828-day-old healthy Arbor Acres broilers were assigned into46treatments randomly, with8replicates each treament (4repeats for male and female respectively), eachrepeat had six chickens. Chickens were sacrificed by using electrical shock after they were feeddifferent semi-pure diets to35day. Ileal digesta were collected to determine the digestibility of aminoacid at the terminal ileal and the concentration of standardized ileal digestible amino acid (SIDAA). Themean values of SID for16kinds of amino acid in corns were as follows: Asp88.56%, Thr81.62%, Ser87.37%, Glu89.24%, Gly82.57%, Ala88.81%, Cys79.92%, Val85.64%, Met92.16%, Ile86.75%,Leu89.30%, Tyr87.68%, Phe88.66%, Lys87.83%, Arg88.90%and Pro84.37%. The mean values ofSIDAA in corns were as follows:0.45%, Thr0.24%, Ser0.36%, Glu1.26%, Gly0.24%, Ala0.51%,Cys0.12%, Val0.28%, Met0.11%, Ile0.19%, Leu0.80%, Tyr0.25%, Phe0.32%, Lys0.20%, Arg0.35%, Pro0.55%.Experiment3Development of NIRS calibration model on SIDAA46powder corns werescanned by near infrared reflectance spectrometer of DA7200, and the information of diffuse reflectancespectroscopy were collected. The NIR calibration model of the SID of16amino acids and the SIDAAwere established using variety of spectral pretreatments and partial least squares. The results showedthat: NIRS can be used to predict the SID of16amino acids and the content of standardized ilealdigestible amino acid in corns. The calibration model’s R2calfor14amino acids of SID were all exceed 0.85except Cys and Arg, while for12standardized ileal digestible amino acids were exceed0.92excluding Cys, Val, Met, Ile andPhe. The range of main factor for16prediction models was4~9, thenumber of samples in the calibration set was greater than54.
