Objective:We attempt to set up a rat model with a syndrome of damp-heat,and then tried to observe serum and tongue mucosa levels of IL-23and IL-17changes and further we study the relationship between the two and syndrome of damp-heat,last study the significance of IL-23and IL-17in the pathogenesis of damp-heat syndrome through the effect of modified huopuxialing decoction in serum and tongue mucosa.Method:First,96rats were randomly divided into there groups:normal group,damp-heat model group and medicine interference group.The damp-heat syndrome rat model was established by high-fat diet,hot and humid environment and microbial infection method of combining multi-factor.Medicine interference group was intervened by the modified HuoPuXiaLing Tang from the first25days. Dosage was divided into two gavage and taken continuously for7days. Rats that divided into4h,24h,48h,72h,four time were sacrificed and drawn.The level of IL-23and IL-17changes in rat serum were detected by ELISA.We take the tongue tissue, and then make pathological sections in normal way,last observe the changes of tissue pathology under light microscope. We detect the expression of IL-23and IL-17in rat tongue mucosa by immunohistochemistry.Results:1The severity of disease classification were the worst in model4h,and decreased gradually with time point,48h and72h groups were significant differences in comparison with the4h group (p<0.05).72h Group and24h groups also have distinct differences (p<0.05).The symptoms of intervention group alleviate than the model group.Both groups had significant differences by nonparameteic test.The symptoms of drug intervention improvemed significa-ntly (p<0.01)2The levels of expression of IL-17, IL-23in serum and tongue mucosa of model group were significantly higher than the normal group in each time point (p<0.01).The levels of IL-23and IL-17were the highest in model4h,and decreased gradually with time point and72h groups of IL-23and IL-17were significant differences in comparison with the4h group (p<0.05),72h Group and24h groups also have distinct differences (p<0.05).The tongue mucosa of IL-17, IL-23expression levels at each time point were relatively have different (p<0.01)In mild,moderate and severe rat model with syndrome of damp-heat,the serum contents of IL-23and IL-17showed a gradual increasing trend with the clinical symptoms (p<0.01) and serum and tongue mucosa content of the two were positively correlated to the severity of syndrome of damp-heat significantly (p<0.01)The expression of IL-23and IL-17in the febrile disease damp-heat syndrome serum and tongue tissue showes a positive correlation (p<0.01)3The levels of expression of IL-17, IL-23in serum and tongue mucosa of medicine interference group were significantly lighter than the model group in each time point (p<0.01).The levels of IL-23and IL-17were the highest in model4h.To48h,the remaining indicators are higher than the normal group except for the serum of IL-23and decreased gradually with time point.48h and72h groups of IL-17were significant differences in comparison with the4h,24h group (p<0.05).Compared with4h,the expression of trends in serum IL-23in24h,48h,72h were respectively different (p<0.05).The tongue mucosa of IL-17, IL-23expression levels at each time point were relatively have different (p<0.01)In mild,moderate and severe rat model with medicine interference group,the serum contents of IL-23and IL-17showed a gradual increasing trend with the clinical symptoms.Severe rat model group of the two were significant differences in comparison with the mild,moderate group(p<0.01,p<0.05). The remaining indicators are positively correlated with the severity of the disease except of serum IL-23(p<0.01, p<0.05).The expression of IL-23and IL-17in the finterference group serum and tongue tissue showes a positive correlation (p<0.01)Conclusion:1IL-23and IL-17maybe participate in the pathogenesis of syndrome of damp-heat,and the two were significantly positively correlated to the severity of syndrome of damp-heat.2The function of IL-23may through effect the levels of11-17expression in the inflammatory response in the damp-heat syndrome.3Modified huopuxialing decoction can ameliorate model rat’s damp-heat syndrome,and decrease the expression the Serum and tongue mucosa of IL-17, IL-23.
