Objective:Research the efficacy of totle Flavonoid of Pteris multifida in benignprostatic hyperplasia(BPH) mice and the possible mechanism, identify its therapeuticaleffect in mice by detection prostate wet weight and prostatic index(PI), furthermore,reveals the possible mechanism by observe the prostate biopsy and the expression ofprostatic hyperplasia-related growth factor through immunohistochemical staining.Methods: Reproduced BPH model in mice by intraperitoneal injection of testosteronepropionate (10mg/kg/d) for31days, after explicitly modeling success of BPH bydetection prostate wet weight and prostatic index, divided the mice into seven groups:control group, model group, positive control groups include finasteride group and prostatgroup, totle Flavonoid of Pteris multifida low dose group, middle dose group and high dosegroup, intragastric administration of drug respectively, once a day for60days. Mice wereexecuted with cervical dislocation24hours after the last administration, isolated theprostate, weighing and calculation of PI, observe the morphological changes at the prostatetissue under the light microscope, detection the expression of vascular endothelial growthfactor (VEGF), basic fibroblast growth factor (bFGF) and transforming growth factorβ1(TGF-β1) in mice prostate by immunohistochemical staining.Result: Intraperitoneal injection of testosterone propionate can reproduce the modelof BPH in mice successfully, the prostate wet weight, prostatic index were significantlyincreased (P<0.05), the glandular epithelium of BPH model changed to high columnar,cells packed tightly, part of the glandular epithelium hyperplasia like papillary and brokeinto the glandular cavity and showed change like pseudostratified, individual cells can beseen two to three oval-shaped nuclear; glandular expanding significantly, the secretion wassignificantly increased in the cavity; surrounding fibrous connective tissue and smoothmuscle thickening and increased. Prostate wet weight and PI of all treated groups weredecreased, the pathological manifestations improved with various degrees, of which theprostate weight and prostatic index in the high dose group of totle Flavonoid of Pteris multifida and prostat group decreased significantly (P<0.05), the pathologicalmanifestations improved most obvious. The immunohistochemical staining showed thattotal flavonoids of Pteris multifida can significant reduce the expression of VEGF whileincrease the expression of TGF-β1(P<0.01) in hyperplasia prostate of mice (P<0.01), bothof which is better than finasteride.Conclusion: This study showed that intraperitoneal injection of testosteronepropionate can replicate of the model of BPH in mice successful. The total flavonoids ofPteris multifida can significantly reduce the prostate wet weight and prostate index, canreduce the number of glands and makes it smaller in the model, restore the normal form ofglandular epithelium, reduce the glandular secretions and interstitial around the gland; itsmechanism may be related to the total flavonoids of Pteris multifida reduce the stimulatinggrowth factor and regulation the signaling networks of growth factor in prostate, inhibitionof proliferation, accordingly have therapeutic effect.
