Objective:To observe the effect of cluster Needling of Scalp point on cognitive function and expression of VEGF in rats with chronic cerebral ischemia,and explore the mechanism.Methods:To make the chronic ischemia model through improved permanent bilateral carotid arteries occlusion. The surviving rats were randomly divided in to three groups:the group of ischemia,the group of cluster Needling,the group of Nimodipine.To check the ability of leaning and Spatial remembering through Morris water labyrinth;To study the morphologic changes with HE staining and the expression of VEGF by immnunohistochemisty method.Results (1) The changes of the ability of leaning and Spatial remembering: The escape latency in each trial of the sham operation group and the blank control group was no significant change(P>0.05).The escape latency in each trial of the ischemia group were longger than the sham operation group(P<0.01).The escape latency in each trial of the cluster Needling group and the Nimodipine group were shorter than the ischemia group(P<0.01) The escape latency in Needling group and the Nimodipine group was not statistically significant differences(P>0.05).(2) The morphological Changes:It was saw that with HE staining, there was a few theVertebral somatic cells of Frontal cortex in the the sham operation group and the blank control group, the arrangement was arrangement compact,the staining was shallow;The staining of four to Multilayers cells in the hippocampus was shallow, the arrangement was arrangement compact,there was no or a few pyknotic cells.the Vertebral somatic cell number of Frontal cortex in the Ischemia group and the significantly reduced, the arrangement was loose-Gap to surrounding cells was obvious, the formwas irregular,the shape was triangular, Cytoplasmic dyeing inequality but the shading deep, free bubble formed;The hippocampus cell number of the Ischemia group significantly reduced, irregular form, the arrangement was loose, and therewere A large become pyknotic cells. It was saw that with HE staining, theVertebral somatic cell number of Frontal cortex in the cluster Needling group and the Nimodipine group was more, the Arrangement wascompact, theFormwas general rules,and there were A few become pyknotic cells;The hippocampus cell number was mild decrease, the form was clear, and there were a few become pyknotic cells.(3) The results of immunohistochemical test: Compared to the blank control group, the expression of VEGF in the cortex of frontal lobe and the hippocampus of rats of the sham operation group was no significant difference(P>0.05);Compared to the sham operation group The expression of VEGF in the cortex of frontal lobe and the hippocampus of rats in other groups wassignificantly more(P<0.01); Compared to the Ischemia group, The expression of VEGF in the cortex of frontal lobe and the hippocampus of rats inthe cluster Needling group and the Nimodipine group was Significantly increased(P<0.01). The expression of VEGF in the cortex of frontal lobe and the hippocampus of rats between the cluster Needling group and the Nimodipine group was not statistically significant differences(P>0.05).Conclusion:1.Successful with2VO method to establish the chronic cerebral ischemia rats model.2,The method of cluster Needling of Scalp point can improve the ability of leaning and Spatial remembering in rats reduced by chronic cerebral ischemia.3. The method of cluster Needling of Scalp point can Obviously increase the expression of VEGF in the cortex of frontal lobe and the hippocampus of rats reducd by chronic cerebral ischemia,It is prompt that the method of cluster Needling of Scalp point can improve the ability of leaning and Spatial remembering in rats reduced by chronic cerebral ischemia through enhacing the expression of VEGF.
