Objective:Epilepsy is a common clinical syndrome in nervous system,which is based on repeated epileptic seizures caused by abnormal discharge ofbrain neurons simultaneously and is sudden, repeated and chronic centralnervous system dysfunction. Recently, Most researches shows that neuralimmune regulatory network imbalance may be involving in the pathogenesisof epilepsy before and after taking antiepileptic drugs. In this experiment,through the detection of the levels of serum immunoglobulins lgA, IgG, IgMand CD3、CD4、CD8、ICAM-1in children who are diagnosed with primaryepilepsy before and after taking antiepileptic drugs,such as LEV and LTG, toinvestigate the changes of immunization levels and the influence of newantiepileptic drugs LEV, LTG on immune ability in children with epilepsy, andto understand the pathogenesis of epilepsy, also the new possible mechanismsof antiepileptic drugs, since it can lead a new approach to treatment.Methods:This study belongs to experimental case-control study.Experimental subjects:epileptic group:50children newly diagnosed primaryepilepsy, who were treated at neurology clinics and wards of Hebei Children’sHospital from September2010to December2011, were chosen as epilepticgroup, they were divided into LEV group and LTG group according to clinicaland electroencephalogram features.Normal control group:another20healthy children from Children Healthcare department of Hebei Children’s Hospital,who were excluded with thedisease of neurous and immunity system by strict physical examination,werechosen as normal control group.All children were collected2ml blood without anticoagulant and1ml withsodium citric acid blood anticoagulant with empty stomach on early morning. The blood samples of epilepsy group were collected respectively before takingmedication,3months and6months after taking medication, while bloodsamples were collected only once from the healthy normal control group.Applying flow cytometry (USA BECKMAN COULTER company, modelEpics XL) to detected levels of CD3, CD4, CD8and adhesion molecule-1(ICAM-1) and automatic biochemical analyzer (Hitachi7060automaticbiochemical analyzer), to detect levels of serum immunoglobulins lgG, IgA,IgM through immune turbidimetry.The statistical analysis SPSS16.0statistics software was used to carry onstatistics processing, p<0.05(double side) had statistical significance.Result:1Compared with the normal control group, the levels of IgA, IgG, ICAM-1,CD8levels increased, and CD3、CD4levels increased in the blood of childrenwith epilepsy before treatment, there was significantly statistical differencebetween them(P<0.05),whereas level of IgM had no statistical differenceswith that of normal control group;2After3months treatment with LEV, IgA、IgM levels decreased, and thelevels of CD8increased,that had Significantly statistical differences with thatof taking mediation, but the increased of CD4、ICAM-1and the decreased ofIgG、CD3compared with before treatment group was no Significantlystatistical difference;After6months treatment with LEV, the levels of IgA、CD8、ICAM-1decrease, and the levels of CD3、CD4increased,which were significantlystatistical difference with before treatment group and the treatment of the6months, while the increases of IgM levels had statistical differences withbefore treatment group;Compared with the normal control group,IgA、CD3、CD4levels decreased,and CD8、ICAM-1levels increased significantly, there was no statisticaldifferences between the normal control group and6months after treatmentwith LEV;3After3months treatment with LTG, levels of IgA、IgG、ICAM-1decreased, there was Significantly statistical difference with before treatment group,while the levels of IgM、CD3、CD4、CD8were no statistical differences withbefore treatment group;After6months treatment with LTG, the levels of IgA、IgG、CD8, ICAM-1decreased, CD4levels increased, there had significantly statistical differencewith before treatment group,but the levels of CD3、 IgM had no statisticaldifferences;Compared with the normal control group, The, decreased of CD3、CD4、CD8after3month and the decreased of CD4、CD8after6month hadsignificantly statistical differences;4Between the antiepileptic drug LEV and LTG, the levels of CD8increasedbefore treatment,the IgG levels decreased after3month had statisticaldifferences with children taking LTG;After6month treatment with LEV,thelevels of CD3、CD4increased,and the levels of CD8decreased,they hadstatistical differences.Conclusion:Cellular immunity and humoral immunity functions weredisordered in children with primary epilepsy before taking antiepileptic drugs,it was the mainly feature that the levels of CD3、CD4decreased and the levelsof IgA、IgG、CD8、ICAM-1increased; The levels of ICAM-1increased inchildren of epilepsy,which was suggested that it was possible inflammatorycytokines involving in the pathogenesis of epilepsy. There was a change inimmune function in children diagnosed with epilepsy,compared with itselfbefore and after taking antiepileptic drug LEV and LTG. The antiepilepticdrugs can not only control the epileptic seizures,but also have a regulate effectin the immune-function of children with epilepsy,such as LEV and LTG. Theycould be able to improve the immunity functions of the epileptic children。There was abnormal immune-function in children with epilepsy, it wassuggested that we should be pay attention to improving the immune functionat the same time of the clinical treatment of the antiepileptic, and to improvethe quality of life of children and improve the prognosis.
