Objective:Observing bladder tissues HE staining, bladder capacity,NGF and its receptor TrKA,and the influence of downstream PI3-K, Akt contents of rats which are neurogenical bladder(NB) urinary retention(UR) after sacral spinal cord injured(SSCI) treat by the electrical acupuncture of CiLiao, ZhongJi, SanYinJiao accupoint,we would explore possible repaired mechanism about the bladder function after SSCI through the three accupoints.Methods:Randomly select10as blank group of40SD female rats. The other30rats are randomly divided into Model group, Duizhaodian group and Acupoint group after made model of UR after SSCI as10a group. When rats safely pass the spinal shock period,give with7times different treatment accord to their group at the the15th day after operation. Testing the maximum bladder capacity of all rats are gived with initiate bladder capacity at the14th and22th day after operation.Putting to all rats to death after7times treatment, Observing the bladder tissue morphology by HE staining,testing NGF and TrKA expression by western blot, testing PI3-k and Akt expression by ELISA.Result:1. The Maximum Bladder Capacity(MBC):①Each group compares with before,The MBC of Duizhaodian group and Acupoint group is lower than before (P<0.05), but The Acupoint group is much lower (P<0.01);②The Duizhaodian group and Acupoint group’s MBC is lower than the Blank group and Model group after treatment,and the MBC difference before and after treatment(d value) is bigger than the Blank group and Model group,which has significant difference (P<0.05);While the MBC of the Acupoint group is much lower and the d value is much bigger than the Duizhaodian group after treatment comparing with the blank group and model group, which has statistically significant (P<0.05).2.HE staining to observe histomorphology of bladder:①The Blank group:Morphology of cells of normal bladder tissues, there was no inflammatory cell infiltration;②The Acupoint group:epithelial tissue levels clear,continuous and surface cells are normal. Few visible cells and neutrophils, abnormal bleeding or capillary expansion can occasional been see between the grass-roots level and pithelial tissue;③The Duizhaodian group: mucosal epithelial tissue level is disorder, the surface of cell sizes and many vacuoles cells, mucous membrane epithelial tissue and muscle layers are seen many neutrophils infiltration, mucosal epithelial tissue and muscle layer more visible abnormal bleeding or expansion of capillary vessels;④The Model group:epithelial tissue level is disorder, epithelial cell falls off, bladder wall thinning, numerous vacuoles cells,mucous membrane and muscle layer is a large number of neutrophils infiltration, and large blood vessels are abnormal bleeding or severe blood capillary expansion.3. Detect the content of NGF in bladder tissues by Westernblot method:The content of NGF of the Model group and Acupuncture group is higher than that in the Blank group with significant difference (P<0.01),and the NGF level of the Acupoint group increased more significantly compared with the Duizhaodian group and the Model group(P<0.05, P<0.01).4. Detect the content of TrKA in bladder tissues by Westernblot method:The content of TrKA of the Model group,Duizhaodian group and Acupuncture group is higher than that in the Blank group with significant difference (P<0.01),and the TrKA level of the Acupoint group increased more significantly compared with the Duizhaodian group and the Model group(P<0.01).5. Detect the content of PI3-K and Akt in bladder tissues by ELISA method:The content of PI3-K and Akt of the Model group,Duizhaodian group and Acupuncture group is higher than that in the Blank group with significant difference (P<0.01),and the PI3-K and Akt level of the Acupoint group increased more significantly compared with the Duizhaodian group and the Model group(P<0.01).Conclusion:1. The rate of spinal cord transection preparation of sacral neurogenic urinary retention after injury in the rat damage model is high. Double lower limbs of rats is typical of a drag, independent maximum bladder capacity increasing, and dysuria and urinary retention.2. Sacral neurogenic bladder of rats injure characterizing by increasing bladder capacity. Giving priority to with urinary retention of bladder dysfunction, and cupping chiliao point, zhongji point, sanyinjiao point to strengthen the detrusor retraction force, it reduces the size of the bladder and reduces the residual urine volume and relieves the symptoms of urinary retention.It is conducive to recovery of bladder function. It also Can alleviate bladder tissue inflammation, alleviate bladder tissue morphological occurrence pathological changes degree.3. Cupping chiliao point, zhongji point, sanyinjiao point can make the model NGF and its receptor TrKA proliferation, and activate downstream of the PI3K/AKT pathway, activate antiapoptotic factors relating to cell apoptosis, inhibit the urinary bladder and promote restoration of the damaged tissue regeneration. Thus delaying the lumbosacral cord injury bladder dysfunction after the urinary retention caused by the secondary injury of the urinary bladder and lead to apoptosis,it plays to protect and repair of damaged bladder tissues and cells, as well as to achieve the aim of repairing the bladder function.
