In this study, the bacteria from the milk of the dairy cows suffering from clinicalmastitis were isolated. The milk was uniformly smeared on the Maconkey agar, the eosinmethylene blue medium and the rabbit blood nutrition agar plates respectively. Thespecific colonies were selected and observed under microscope. The colonies suspected asE. coli were inoculated on the eosin methylene blue agar plate and the suspectedStaphylococcus aureus and Streptococcus on the rabbit blood nutrition agar platesrespectively. The purified bacteria were identified by Biochemical tests and PCR. Itfound that E. Coli, Staphylococcus aureus and Streptococcus were the main pathogens ofthe milk.15kinds of Chinese herbal medicine such as Scutellaria baicalensis Georgi, Leonurusjaponicus Houtt, Lonicera hypoglaucaMiq, etc. were used to study the antibiosis effectsof the3main pathogens on mastitis. It found that Staphylococcus aureus was highlysensitive to Scutellaria baicalensis Georgi, Sanguisorba officinalis L., Menthahaplocalyx Briq, Taraxacum mongolicum Hand-Mazz. and moderately sensitive toForsythia susupensa(Thunb.)Vahl., Angelica sinensis (Oliv.) Diels,Pulsatilla chinensis,Angelica dahurica, Viola yedoensis Makin. E.coli. was moderately sensitive toScutellaria baicalensis Georgi, Forsythia Susupensa (Thunb.) Vahl. Taraxacummongolicum Hand-Mazz., and Sanguisorba officinalis L. And Streptococcus agalactiaewas moderately sensitive to Sanguisorba officinalis L., Scutellaria baicalensis Georgi,Pulsatilla chinensis, Forsythia susupensa (Thunb.) Vahl., Angelica sinensis(Oliv.)Diels,Taraxacum mongolicum Hand-Mazz., Angelica dahurica and Mentha haplocalyx Briq.According to the antibacterial trial results, Scutellaria baicalensis Georgi, Leonurusjaponicus Houtt, Lonicera hypoglaucaMiq, Forsythia susupensa(Thunb.)Vahl., andSanguisorba officinalis L. were chosen to form6different optional prescriptions on thebasis of the principle of "Clearing Heat, relieving toxicity and promoting bloodcirculation " in traditional Chinese Veterinary medicine. Mouse ear edema model andmouse peritoneal inflammation model were induced by acetic acid to study theanti-inflammatory effects of different formulas. It showed that Prescription Two andPrescription Three had significantly anti-inflammatory effects. Prescription Three wasconfirmed as the ultimate prescription and made perfusion agent.In order to evaluate the safety of the Chinese medicinal herbal compound filler, theacute dermal toxicity, the skin irritation and the skin allergy trials were carried out. Itdemonstrated that the Chinese herbal compound perfusion agent had no toxicity in theacute dermal toxicity trial. The stimulation index was less than0.5, the allergy index was0. It showed that the perfusion agent was safe. To investigate the clinical effects of Chinese medicinal herbal compound filler,theclinical mastitis model of dairy goat was made by the3main pathogens infection. After5days of therapy, the nipple swelling of the dairy goats significantly reduced, the colorof the milk turned lighter and the somatic cells were significantly fewer. After thetreatment, the bioactivity of SOD and GSH-Px in the sera increased significantly, thelevel of NO, MDA significantly declined. The results indicated that the therapeuticeffects of Chinese herbal medicine on dairy cow mastitis related to its action onenhancing body’s antioxidant capacity and reducing the releasing of inflammatoryfactors.
