The"Rukangsan"used in treating mastitis of dairy cows was screend through Chinese veterinarian theory and clinical experiment. Then evaluated the safety, anti-inflammatory andclinical effect of"Rukangsan", at last tested the efficacy mechanism of the medicine.1. Antibacterial test of 16 kinds of Chinese herbal medicines and component medicines to the 3 kinds of main pathogens on cow mastitis The antibacterial test of 12 kinds of single herbs and 4 kinds of antibiotics to the 3 kinds of main pathogens commenly caused mastitis of dairy cows was done, the result indicated that the inhibitory effect of berberine,dandelion were better, and lactic acid ciprofloxacin was the highest sensitivity. According to the single herbs antibacterial effect, antibacterial tests of 4 different components of compound recipe were designed; the results indicated that compound recipe 2 was rather better in inhibitory effect. This test provided a scientific basis for designing of compound drugs.2. The screening experiment on the"Rukangsan"formulaThe 183 cows were treated for clinical mastitis test by three kinds of traditional Chinese medicine.It will screen the best of compound results were as follows: acute mastitis cure rates on compound1, compound 2, compound 3 were 61.5%,82.7%,54.2% respectly; The cure rates on subclinical mastitis were 70.3%, 96.0%, 95.2% respectly. By statistical teste, the effects of acute mastitis of compound 2 was the best, compound 2 significantly different than compound 3 (P <0.05); compound 2, Compound 3 were the best to subclinical mastitis,they compares with compound 1 significant difference (P<0.05). The compound 2 was screened as the best prescription.3. Safety evaluation on"Rukangsan""Rukangsan"safe was trialed .The mice were animal model.Acute toxicity test was did to determine its half leathal dose. The results: mice did not die, LD50>60000mg/kg."Rukangsan"was safe and reliable.It would provide a theoretical basis for clinical application.4. The anti-inflammatory test on"Rukangsan"According to the Chinese veterinary basic theory, developed a pure Chinese medicine"Rukangsan"on treatment of cow mastitis, prepare for the anti-inflammatory effects of the drug to mice as animal models, designed two groups of different types of tests. The results showed that high(20g/kg),medium(10g/kg) and low(5g/kg) concentrations of Chinese medicine compound preparation could significantly reduce the rate of mice ear swelling(P<0.05).The swelling level were 10.23mg, 8.81mg, 8.43mg and 12.42mg respectly; cotton-granulation tissue proliferation (P < 0.05) It was 10.92mg,8.72mg,10.43mg and12.54mg respectly.The result showed that the compound preparation of traditional Chinese medicine can effectively fight against different types of acute and chronic inflammation; anti-inflammatory effect significant for clinical application provides a theoretical basis.5."Rukangsan"treatment of acute mastitis in dairy cow observation 146 dairy cows of acute mastitis were treatment with"Rukansan"(test group),Penicillin,Streptomycin and Gongyinsan(control group).It was found that Rukansan, Penicillin,Streptomycin and Gongyinsan on dairy acute mastitis cure rate 86.5%,66.7% and 60.9% respectively.in the 1 courses (3days) the cure rate were 48.9%,20.0%and 23.9%. After statistical testing, test group and control group cure rate of treatment and there were significant differences on milk powder and Culture short course of treatment, good clinical efficacy.6."Rukangsan"treatment of subclinical mastitis in dairy cow observation100 cows of acute mastitis were treatment with Rukansan (test group), Levamisole(control group).It was found that"Rukansan", Levamisole on dairy Subclinical mastitis cure rate 94.6% and 46.5% respectively. Rukangsan increased significantly milk production.After statistical testing, test group and control group cure rate of treatment were significant difference.7.Effects of"Rukangsan"on the albumin,relationenzyme in the milkTaking the Penicillin and Streptomycin as the comparison, the curative effect of Rukangsan was observed for clinical dairy cows with acute mastitis. Lactate dehydrogenase (LDH),glutamlc pyruvic transam Jnase(GPT),rum albumin (SA) were detected in milk . Result: the acute mastitis's cure rate on Rukangsan,Penicillin and Streptomycin was significantly different, they were 2.62 g/L,2.65 U/L,21.13 U/L respectively; Rukangsan may drop SA, GPT, and LDH to the normal level in 3rd day or 7th day. It was significantly difference than 0 day. Penicillin and Streptomycin may drop SA, GPT to the normal level in 7th day, they were 2.41 g/L,2.63 U/L respectively,but it had the fluctuation. It would elevate in 15th day. LDH was reduced, but could not fall to the normal level.8. Effects of"Rukangsan"on the the total number of bacteria albuminin in the milkThe total number of bacteria was detected When"Rukangsan"casual treatment of the milk of cows. The Green streptomycin was control group. Using colony counting assay plate milk detected total bacteria. The results showed that: "RukangSan" group and Penicillin,Streptomycin group ,the total number of bacteria were 160,000 / mL, 210,000/ mL,at the 7th day respectively."RukangSan"group compared with the healthy group significantly different, with Penicillin,Streptomycin group no significant difference; Penicillin,Streptomycin group in the first 15 days increased the total number of bacterial streptomycin group health, compared with significant difference (P <0.05), " RukangSan " group did not increase.
