Objective:To observe the immunogenicity of pepsin deproteini-zed xenogeneic bone graft material for repairing bone defect of rabbit radius.Method:125New Zealand rabbits were randomly divided into5groups, including blank group, pepsin group, autogenous bone group, bone matrix group, hydrogen peroxide group. Each group had25rabbits. Operation caused rabbit left forelimb segment bone defect of radius10mm. The blank group didn’t implant any material in bone defect. Other groups were implanted with autogenous bone, bone matrix, hydrogen peroxide or pepsin deproteinized xenogeneic bone respectively. Postoperative3d,7d,14d,28d,56d, each group were sacrificed5rabbits. Appropriate specimens were detected by flow cytometry and RT-PCR, ELISA and histological observation.Results:The postoperative flow cytometry, RT-PCR and ELISA results showed autogenous bone group and blank group indicators were lower than the other groups, the indicators of pepsin group and bone matrix group were lower than hydrogen peroxide group, and there is no statistically significant differences between pepsin group and bone matrix group; histological observation showed autogenous bone group had few inflammatory cells and lymphocytes, pepsin group and bone matrix group had less inflammatory cells and lymphocytes than hydrogen peroxide group.Conclusion:The immunogenicity of the pepsin group and the bone matrix group was less than hydrogen peroxide group. The immunogenicity of the pepsin group and the bone matrix group. There is no significant difference of immunogenicity between pepsin xenogeneic bone graft material and medical induced bone matrix. The pepsin xenogeneic bone graft material reached a preliminary requirements for clinical bone graft substitute material.
