| Objiects:To investigate the therapeutic effects of Novaferon combined with mesalazine and Prednisone on ulcerative colitis.Methods:We used4%-DSS-induced acute colitis model to investigate the therapeutic effects of Novaferon.70mice were randomly divided into seven groups as follow:normal control group,model control group,mesalazine group,prednisone group,low-dose novaferon group,moderate-dose novaferon group,high-dose novaferon group.The normal group were only fed with distilled water.The other group were fed with4%dextran sulfate sodium(DSS) for7consecutive days.Starting from the next day,besides the normal control group and the model control group,each group was given low-dose novaferon, moderate-dose novaferon, high-dose novaferon, mesalazine and prednisone. The normal control group and the model control group were given NS to insert stomach.After8days,all mice were sacrificed. Severity of colitis was evaluated by disease activity index (DAI), colon length, histology, and pronflammatory cytokine production including tumor necrosis factor-a (TNF-a) and interleukin-5(IL-5). Results:1.Novaferon can reduce the DAI and histology scores of mice with ulcerative colitis.2.The expression of IL-5and TNF-a was increased(p<0.05) in the mice which was in the group of low-dose novaferon,moderate-dose novaferon,high-dose novaferon,mesalazine and prednisone combined with the normal control group.The expression of IL-5was decreased(p<0.05) in the mice which was in the group of low-dose novaferon,moderate-dose novaferon,high-dose novaferon,mesalazine and prednisone combined with the model control group.The expression of TNF-a was decreased(p<0.05) in the mice which was in the group of high-dose novaferon,mesalazine and prednisone combined with the model control group.Conclusion:1.Novaferon can reduce the DAI scores and tissue damage of mice with experimental colitis2.Novaferon can reduce the expression of IL-5and TNF-a in intestinal mucosa of mice with experimental colitis... |
