Objective:TO study the mRNA expression of MMP1and DnaselL3in the oral verrucous carcinoma(OVC) and oral squamous cell carcinoma andnormal paracancerous and normal oral mucosa tissue. Preliminary validation the accuracy of gene chip. To study the expression of MMP1and Dnase1L3in the oral verrucous carcinoma, oral squamous cell carcinoma andnormal oral mucosa tissues by immunohistochemistry and westernblot. Investigating the role in the development process for MMP1and Dnase1L3in the oral verrucous carcinoma and proving the profit for early diognosis,prognosis assessment and treatment of OVC.Methods:(1) We collected the tissue of oral verrucous carcinoma、 oral squamous cell carcinoma andnormal and normalmucosa from5patients to study the expression of MMP1and Dnase1L3genes at mRNA level by using RealTime-PCR.(2) Using immunohistochemical technique, we uncovered the differential expression of MMP1and DnaselL3protein in17patients of oral verrucous carcinoma17patients of squamous carcinoma and17of normal mucosa tissue.(3)Using WseternBlot technique,we uncovered the differential expression of MMP1and Dnase1L3protein in3patients of oral verrucous carcinoma3patients of squamous carcinoma and3of normal mucosa tissue. Results:Expression of MMP1mRNA in OVC-CA was significantly higher when compared to paracancerous(OVC—P)and normal oral mucosa tissues(OVC-N)(P<0.05). There was not statistically significant(P>0.05) in the expression of MMP1mRNA in the tissues of OVC-CA and OVC-P、OVC-P and OVC-N. Expression of Dnase1L3mRNA OVC-CA was significantly lower when compared to paracancerous(OVC-P)and normal oral mucosa tissues(OVC-N)(P<0.05). But There was not statistically significant(P>0.05. Expression of MMP1mRNA OSCC-CA was significantly higher when compared to paracancerous(OSCC-P)and normal oral mucosa tissues(OSCC-N)(P<0.05). Expression of DnaselL3mRNA OSCC-CA was significantly lower when compared to paracancerous(OSCC—P)and normal oral mucosa tissues(OSCC-N)(P<O.05)。But there was not statistically significant(P>O.05) in the expression of MMP1mRNA in the tissues of OSCC-CA and OSCC-P、OSCC-P and OSCC-N.The expression of MMP1of OVC、OSCC、NM was significantly different when totally compared(P<0.05), Expression of DnaselL3protein in OVC and OSCC was significantly lower when compared to the normal oral mucosaConclusions:1. The expression of MMP1mRNA Was at high level in oral squamous cell carcinoma andnormal, There Was higher significant differencof MMP1expression between OSCC—CA、OSCC-P and OSCC-N. the expression of DnaselL3mRNA Was at low level in oral squamous cell carcinoma andnormal。 But there was significant difference between OSCC—CA and OSCC-N, Thus MMP1mRNA might be a functional factor in the initiation and development of OSCC.2. The expression of MMP1mRNA Was at high level at the mRNA and protein level in the OVC, The result Was as same as the results of previous gene chips. Thus MMP1mRNA might be a functional factor in the initiation and development of OVC.3. MMP1can be a used as the prognosis factors of oral verrucous carcinoma and oral squamous cell carcinoma andnormal, there may be in the gene leve to lthrough MMP1inhibitors to treatment from oral squamous cell carcinoma and verrucous carcinoma purpose.
