Objective: To study the relationship the allele frequencies and genotypedistribution of interleukin-18(IL-18)gene polymorphism patients with in type2diabetes mellitus and diabetic nephropathy (DN),And to analyze association theserum levels and genotype of IL-18with type2diabetic nephropathy.Methods: The polymorphisms of IL-18gene, including polymorphisms ofIL-18gene promoter-137G/C and-607C/A were analyzed by the polymerasechain reaction-restriction fragment length polymorphism (PCR-RFLP) method in360patients with type2diabetes mellitus and180healthy controls, and the serumlevel of IL-18was determined by enzyme-linked immunosorbent assay(ELISA).Automated Biochemical Analyzer was used to measure the serum totalcholesterol(TC), triglyceride(TG), high density lipoprotein cholesterol(HDL-C),low density lipoprotein cholesterol (LDL-C), apolipoprotein AI(apoAI) andapolipoprotein B(apoB). fasting plasma glucose(FPG), were measured.Results:(1)IL-18gene-137C/G polymorphism CC,GC,GG genotypes wasin Hans population of Guangxi-guibei,the frequency of IL-18GG genotype wasthe highest among IL-18genotypes (74.4.7%),GC+GG genotype(25.6%),andthese gene polymorphisms had no difference between male and female (P0.05). IL-18gene-607C/A polymorphism CC,GA,AA genotypes was in Hanspopulation of Guangxi-guibei,the frequency of IL-18CA genotype was thehighest among IL-18genotypes (51.9%),GC+GG genotype(48.1%),and thesegene polymorphisms had no difference between male and female (P0.05).(2)Thediabetic nephropathy group showed significantly higher serum levels of IL-18than control group (P<0.01).(3)The distributions of IL-18gene-607C/Apolymorphism was not different between type2diabetic nephropathy group andcontrol group, but the IL-18gene-137G/C polymorphism was significantlydifferent. The relative risk suffered from type2diabetic nephropathy of C allelewas1.876times of the G allele(OR=1.876,95%CI:1.259~2.795), the serumlevel of IL-18C allele carriers was significantly higher than no carriers(77.30±25.33pg/ml VS65.28±22.16) pg/ml P<0.05).Conclusions: IL-18gene-137G/C polymorphism was associated with type2diabetic nephropathy, and C allele may be a risk factor for type2diabeticnephropathy. In which the IL-18C allele carriers may be increase risk byenhancing the IL-18expression in the pathogenesis of type2diabeticnephropathy.
