Expression Of CD4+CD25+Regulatory T Cells In Non-neoplastic Epithelial Disorders Of The Vulva

Objective:To investigate the correlation between the expression level and the incidence of CD4+CD25+Treg cells in lesion tissue and peripheral blood of patients with non-neoplastic epithelial disorders of the v ulvar(NNEDV).Methods:Select30patients with non-neoplastic epithelial disorders of vulva (15cases of vulvar squamous hyperplasia,15cases of vulvar lichen sclerosus) as the experimental group, and5cases of myoma of uterus or uterine prolapse as the control group. Detect the differences of vulva skin protein expression in CD25+T cells between the experimental group and the control group by immunohistochemical method (Immunohistochemistry IHC); Detect the expression of CD4+CD25+Treg cells in peripheral blood between the experimental group and the control group by flow cytometry.Results:1. the result of immunohistochemistry showed that CD25protein was mainly located in cytoplasm and cell membrane of lymphocytes. The expression of CD25+T cells in the skin lesions of non-neoplastic epithelial disorders of vulva patients was higher than normal control group (p=0.000); The expression of CD25+T cells in the skin lesions of lichen sclerosus vulvar squamous epithelial hyperplasia patients was higher than vulvar squamous hyperplasia patients(p=0.000);2. The result of flow cytometry showed that the expression of CD4+CD25+Treg cells in peripheral blood of non-neoplastic epithelial disorders of vulva patients was higher than that of normal control group (p=0.000), and the expression of CD4+CD25+Treg cells in peripheral blood of lichen sclerosus vulvar squamous epithelial hyperplasia patients was higher than vulvar squamous hyperplasia patients(p=0.000). Conclusion:1. The increasing number of CD4+CD25+Treg cells may be involved in the incidence of non-neoplastic epithelial disorders of vulva;2. Non-neoplastic epithelial disorders of vulva may exist immune disorders, and the immune disorder in vulvar lichen sclerosus is more serious vulvar squamous epithelial hyperplasia.
